摘  要:目的 观察表皮生长因子受体(EGFR)内源性抑制因子LRIG1在转录水平于星形细胞瘤中的表达及其与EGFR mRNA和肿瘤恶性程度的相关性,探讨其在肿瘤发生中的作用.方法 逆转录-聚合酶链反应(RT-PCR)法检测LRIG1和EGFR mRNA在35例Ⅰ~Ⅳ级星形细胞瘤组织中的表达水平.统计学分析各组间表达的差异性.结果 在多数肿瘤组织中LRIG1 mRNA低表达(27/35),EGFR mRNA存在过度表达(21/35),在18例低级别(Ⅰ~Ⅱ级)与17例高级别(Ⅲ~Ⅳ级)肿瘤中前者表达差异无统计学意义(P〉0.05),后者表达差异有统计学意义(P〈0.05).两者表达趋势无明显相关(r=0.187,P〉0.05).结论 作为EGFR信号系统的负反馈调节因子,LRIG1在mRNA水平的表达下调提示了它和星形细胞瘤的发生有关,其与EGFR的表达趋势及肿瘤恶性度的非相关提示其可能作为胶质瘤发生的早期影响因素.Objective To evaluate the expression of human leucine-rich repeats and immunoglobulin like domainsl (LRIG1) mRNA in astrocytoma and its correlation with epidermal growth factor receptor (EGFR) mRNA and tumor malignancy in order to investigate its role of LRIG1 in the oncogenesis. Methods The expression of LRIG1 and EGFR mRNA in 35 cases of astrocytomas with grades I-IV was detected by RT-PCR,and analyzed statitiscally. Results There was weak expression of LRIG1 mRNA (27/35) ,and over-expression of EGFR mRNA (21/35) in most astrocytomas. There was no significant difference in the LRIG1 mRNA expression between low grade (Ⅰ~Ⅲ) (18 cases) and high grade (Ⅲ~Ⅳ) (17 cases) astrocytomas (P 〉 0. 05) , but there was significant difference in the EGFR mRNA expression between them. There was no significant correlation between the expression of LRIG1 mRNA and EGFR mRNA (r = 0. 187, P 〉 0.05). Conclusion As a feedback negative attenuator of EGFR signaling, the LRIG1 mRNA was down-regulated, suggesting its contribution to the oncogenesis of astrotoma. However, there was no significant correlation of the LRIG1 mRNA with EGFR mRNA and tumor malignancy, indicating LRIG1 might be the early influencing factors responsible for the pathogenesis of astrocytoma.
