云南肺癌患者HLA-A,B基因多态性分析

摘　　要：目的测定云南肺癌患者HLA-A,B等位基因出现频率,探讨我省肺癌发病与HLA-I类等位基因位点的相关性,为肺癌易感的相关机制研究提供有意义的群体遗传资料.方法应用病例-对照相关分析方法,采用PCR-SSP(序列特异性引物基因分型技术)选取籍贯云南的30例云南肺癌患者和30例健康个体(均为随机抽样且无血缘关系),测定其HLA-A,B等位基因出现频率,将其结果与同地区正常组进行比对,计算相对危险因子(relativerisk,RR).对正、负相关值以内的等位基因遗传因素和临床性状数据进行相关分析.结果肺癌组HLA-A*02(90%)(以A*0201为主),B*46(40%)的频率明显高于对照组HLA-A*02(43.3%),B*46(0%),Pc<0.05,RR>1;肺癌组A*31(3.3%),A*33(6.7%),B*27(3.3%),B*52(6.7%)的频率则明显低于对照组A*31(23.3%),A*33(26.7%),B*27(26.7%),B*52(26.7%),Pc<0.05,RR<1.结论云南肺癌患者HLA-A,B等位基因中以HLA-A*02出现频率尤高(90%),可能与云南肺癌易感相关;HLA-A*31,A*33,B*27,B*52的超低频出现提示在云南肺癌发病中有保护作用.Objective To evaluate the frequency of HLA-A,B genotype in patients with lung cancer in Yunnan and study the correlation between lung cancer and HLA-A,B genotype,which may provide significant genetic information for lung cancer research.Methods The HLA-A,B alleles were typed by PCR-SSP assay in 30 Yunnan lung cancer patients and 30 healthy subjects.We compared HLA-A,B frequency between patients and healthy subjects and calculated the relative risk factor.Results The frequency of HLA-A02（90%）,especially A0201,B46（40%）,in the lung cancer patients was significantly higher than that in healthy subjects HLA-A02（43.3%）,B46（0%）,Pc0.05,RR1.The frequency of HLA-A31（3.3%）,A33（6.7%）,B27（3.3%）,B52（90%） in the lung cancer patients was significantly lower than that in healthy subjects A31（23.3%）,A33（26.7%）,B27（26.7%）,B52（26.7%）,Pc0.05,RR1.Conclusion The significantly higher frequency of HLA-A02（90%） in patients with lung cancer in Yunnanmay be associated with the susceptibility to the lung cancer.The extra low frequency of A31,A33,B27,B52 may have protection against lung cancer.
