摘 要:目的研究不同浓度格列卫对恶性淋巴瘤细胞株Raji细胞的影响.方法采用MTT法检测不同浓度格列卫对Raji细胞在不同时间的生长抑制作用,应用流式细胞术检测药物对Raji细胞Caspase-3的表达及G1期、S期占整个细胞周期的比例,采用免疫组化SABC法检测药物对Raji细胞P16蛋白的表达.结果格列卫使细胞存活率明显下降(P<0.05);未能上调Caspase-3表达(P>0.05);无明显诱导凋亡作用(P>0.05);格列卫在低浓度72 h与高浓度48 h、72 h时P16蛋白表达与对照组相比有显著差异(P<0.05);格列卫对Raji细胞G1期、S期所占细胞周期的比例与对照组相比无显著差异(P>0.05).结论STI571在高浓度时可以上调Raji细胞P16蛋白的表达;STI571对恶性淋巴瘤细胞株Raji细胞无上调Caspase-3表达及影响细胞周期的作用.Objective To investigate the effect of gleevec with different concentration on Raji cells and the mechanisms Methods The inhibition of cell proliferation was measured by MTT assay to assess the cells survival after ST1571 treatment with different concentration at indicated time. Caspase-3 activeness changes were determined by flow cytometry (FCM) after treatment with ST1571 in different concentration at indicated time. The relative number of cells in different phases and the percentages of cells calculated in G1 and S phase of the cell cycle were assessed by FCM after treatment with STI571 in different concentration at indicated time. The expression of p16 protein was analysed by SABC immunohistochemical method after treatment with ST1571 in different concentration at indicated time. Results The results of MTT assay showed that STI571 could inhibit Raji cells growth. There was clear statistical significance between the union groups and the single-drug group (P〈0.05). Raji cell lines were less sensitive to gleevec (P〉0.05). At higher concentration and longer time,ST1571 could up-regulated the optical density of P16 protein. And it was found that ST1571 had no effect on Raji cell cycle. (P〉0.05). Conclusions The results showed that ST1571 might affect the expression of Pl6 protein at higher concentration.
