依地酸二钠-几丁糖凝胶对兔角膜血染铁离子析出的作用

摘　　要：目的研究依地酸二钠-几丁糖凝胶对体外角膜血染铁离子析出的作用。方法12只(24跟)健康新西兰大白兔,制作角膜血染模型后随机分为3组:生理盐水组、0.37%依地酸二钠(EDTA)组及0.37%EDTA-1.0%几丁糖组,每组8只角膜。将血染角膜平均分为两半,取一半角膜在各组浸泡前及浸泡35 d后分别行Perl’s染色组织病理学观察,了解相应时间点血染角膜中铁离子分布情况。取另一半角膜分别浸泡在3组液体中,每5 d取浸泡液测量铁离子析出浓度,共7次。结果0.37%EDTA-1.0%几丁糖组浸泡液中铁离子析出量在各时间点明显高于其他两组(P<0.001),且浸泡35 d后的角膜上皮与其他两组相比仍较完整,基质水肿较轻,胶原板层排列仍较紧密。结论0.37%EDTA-1.0%几丁糖凝胶在体外促血染角膜中的铁离子析出作用明显优于生理盐水及0.37%EDTA滴眼液,且对角膜组织的结构无明显影响,有望用于临床上治疗角膜血染。Objective To study the iron-ion-chelating effects of 0. 37% EDTA - 1.0% chitosan gel on corneal bllod staining in vitro. Methods Corneal blood staining model of 24 corneas of 12 New Zeal white rabbits were made. The 24 blood stained corneas were divided into 3 groups and then divide equally each cornea for two parts：one part of corneas were put in saline group,0.37% ED- TA eye droup group and EDTA ehitosan gel group 3 ml for 35 days （ take out 0.3 ml every 5 days for the measure of concentration of iron-ion） ; then take corneas soaked 35 days and the other part of corneas before immersing for staining with Perl＇ s for pathological examination, in order to know how the iron-ion is distributed respectively. Results Every sampling of EDTA chitosan gel group chelated markedly higher iron-ion cpmcemtratopm than the other two groups （ P 〈 0. 001 ）, and the corneas of EDTA chitosan gel group had more integrated epithelium, more compact collagen layers and less oedame than the other two gorups. Conclusion 0.37% EDTA -1.0% chitosan gel has the best effect on chelating iron-ion of corneal blood staining among the three groups, and had no distinct effect on the corneas. It＇ s possible that it will be applied to therapy for corneal bllod staining.
