摘 要:目的:研究谷氨酰胺(Gln)联合5-氟尿嘧啶(5-FU)和环磷酰胺(CTX)对人胃癌SGC7901细胞周期和凋亡率的影响。方法:体外培养胃癌SGC7901细胞株,加入Gln及不同浓度的5-FU和CTX后收集细胞。应用流式细胞术检测胃癌细胞周期及凋亡率的变化。结果:5-FU加Gln时胃癌SGC7901细胞凋亡率不同,浓度由0mg/L增加到240mg/L时呈正直线相关(r=0.806,P<0.01),凋亡率随浓度增加而增加。CTX加Gln对胃癌细胞周期及凋亡率无显著影响(P>0.05)。5-FU加Gln组G0/G1期细胞所占比例较CTX加Gln组明显增多(P<0.05)。G2期5-FU加Gln组明显比CTX加Gln组低(P<0.05)。S期两组比较,差异无统计学意义(P>0.05)。结论:Gln和5-FU影响胃癌细胞株的细胞周期和凋亡率。Gln和CTX对人胃癌细胞株的细胞周期和凋亡率均无明显影响。Objective:To evaluate the effect of combination of Gln and 5-FU and CTX on the human gastric carcinoma cell line SGC7901 in vitro. Methods:Different concentrations of 5-FU or CTX and Gln were added in the cultured human gastric carcinoma cell line SGC7901 in vitro. The changes of apoptosis rate and cell cycle were detected by the flow cy-tometry analysis. Results:Combination of Gln and 5-FU could inhibit the cell growth and enhanced apoptosis rate. The concentration of 5-FU and the apoptosis rate of gastric carcinoma cell line SGC7901 had positive linear correlation (r=0.806,P0.01),the apoptosis rate increased with the increase of concentration. There was no change of apoptosis rate and cell cycle by combination of CTX and Gln (P0.05). The rate of G0/G1 phase in cell cycle of 5-FU and Gln group was more than that of CTX and Gln group (P0.05). The rate of G2 phase in cell cycle of 5-FU and Gln group was less than that of CTX and Gln group (P0.05). The rate of S-phase in cell cycle had no difference in two groups (P0.05). Conclusion:Combination of Gln and 5-FU can inhibit the cell growth and enhance apoptosis rate. There is no change of apoptosis rate and cell cycle by combination of CTX and Gln.
