摘  要:目的:探讨小檗碱治疗腹泻的可能分子机制。方法:利用番泻苷A诱导建立BALB/c小鼠的腹泻模型,给予小檗碱治疗处理,采用半定量RT-PCR检测影响肠管钠水吸收的钠-氢交换体1(NHE1)和钠-氢交换体3(NHE3)的表达情况;利用免疫组织化学的方法,作进一步证实。结果:半定量RT-PCR表明小檗碱处理组NHE3mRNA的表达水平较腹泻组小鼠明显增高,但与正常对照组相比,无显著性差异;而NHE1mRNA的表达水平在三组间并无明显差异。免疫组化的结果也证实,与正常对照组和处理组小鼠相比,腹泻组小鼠NHE3的表达水平明显下调。结论:小檗碱可通过上调NHE3mRNA和蛋白的表达而不是NHE1的表达,进而增强肠管对钠水的吸收,从而发挥治疗腹泻的作用。Objective:To explore the molecular mechanisms by which berberine might exhibit its anti-diarrhea effect. Methods:Diarrhea mice model were induced by sennosideaA,and then treated with berberine. The expression of NHE1 and NHE3 mRNA which affect the absorption of Na+ and water was detected by semi RT-PCR.Furthermore,immunohistochemistry was adopted to confirm the results acquired by RT-PCR. Results:Compared with treatment group and control group,the expression of NHE3 were detected by semi RT-PCR and immunohistochemistry were significantly decreased in diarrhea mice induced by sennosideA,but the expression of NHE1 were no statistical significance among there groups. Conclusion:Berberine can increase the expression of NHE3 not NHE1,suggesting berberine might exhibit its anti-diarrhea effect partially by enhancing the absorption of Na+ and water.
