摘  要:目的:观察天麻素(GAS)对百草枯(PQ)和代森锰(MB)致小鼠黑质多巴胺(DA)能神经元损伤的神经保护作用及其机制。方法:用PQ和MB建立C57BL小鼠PD模型,设立空白组、模型组、GAS低剂量组、GAS中剂量组、GAS高剂量组5组,每组15只,在应用PQ及MB后给予GAS干预,观察小鼠行为学改变,进行TH免疫组织化学染色,并测定黑质区MDA、GSH-Px、GSH、SOD的含量。结果:小鼠注射PQ和MB后出现震颤、竖尾、竖毛、运动迟缓、步态不稳、肢体僵硬等症状。同时黑质致密部(SNc)的TH阳性细胞明显减少;GAS低、中、高剂量组症状较模型组轻,TH阳性细胞数均较模型组明显增加(P<0.05)。GAS低、中、高剂量组MDA含量(分别为6.5 nmol/mg±2.7 nmol/mg、6.2 nmol/mg±2.3 nmol/mg、5.3 nmol/mg±1.6 nmol/mg)与模型组MDA含量为9.7 nmol/mg±3.1 nmol/mg相比明显降低(P<0.05),GAS低、中、高剂量组GSH-Px、GSH、SOD的含量(GSH-Px的含量分别为3.1nU/g±0.3 nU/g、3.5 nU/g±0.5 nU/g、3.9 nU/g±0.4 nU/g;GSH的含量分别为6.1 mg/g±2.1 mg/g、6.5 mg/g±1.9 mg/g、6.8 mg/g±1.7 mg/g;SOD的含量分别为160.3 nU/mg±7.3 nU/mg、164.5 nU/mg±10.2 nU/mg、182.1 nU/mg±9.7 nU/mg)与模型组GSH-Px、GSH、SOD的含量(2.5 nU/g±0.4 nU/g、4.4mg/g±1.5 mg/g、138.5 nU/mg±6.4 nU/mg)相比明显增高(P<0.05)。结论:GAS对PQ和MB诱导的DA能神经元损伤具有一定的保护作用,其机制可能与清除过氧化脂质及自由基,提高机体抗氧化能力有关。Objective:To study the neuroprotective effects and the mechanism of Gastrodin(GAS) on Paraquat(PQ) and Maneb(MB) induced Parkinson's disease(PD) in C57BL mice.Methods: The PD models were formed with intraperitoneal injections of PQ and MB,divided into 5 groups at random,15 each group: control group,model group,GAS high group,GAS media group,GAS low group,respectively.GAS was administered to mice posterior to PQ and MB.We observed the effects of PQ and MB on the behavioral exhibition,performed TH immunohistochemical staining and characterized the contents of MDA,GSH-Px,GSH and SOD.Results: After PQ and MB injection the mice behaved with tremor,erect tail,piloerection,bradykinesia,instability of gait and hypokinesia.Meanwhile the number of TH positive cells in the SNc decreased obviously.However,compared with the model group,the symptoms in the GAS high group,GAS media group,GAS low group were relieved,the number of TH positive cells in the three groups increased obviously(P0.05).The contents of DMA in the later three groups,which were(6.5±2.7) nmol/mg,(6.2±2.3) nmol/mg,(5.3±1.6) nmol/mg,respectively,dereased obviosly and had the significant difference(P0.05),compared with that in model group,which was(9.7±3.1) nmol/mg.The contents of GSH-Px,GSH and SOD in the later three groups increaed obviosly compared with that in model group,which was(2.5±0.4) nU/g,(4.4±1.5) mg/g,(138.5±6.4) nU/mg,respectively.The contents of GSH-Px in the later three groups were(3.1±0.3) nU/g,(3.5±0.5) nU/g,(3.9±0.4) nU/g,respectively.The contents of GSH in the later three groups were(6.1±2.1) mg/g,(6.5±1.9) mg/g,(6.8±1.7) mg/g,respectively.The contents of SOD in the later three groups were(160.3±7.3) nU/mg,(164.5±10.2) nU/mg,(182.1±9.7) nU/mg,respectively.Conclusion : GAS could protect the dopaminergic neurons damaged by PQ and MB in C57BL mice.The mechanism of GAS in Neuroprotective was probable relevant to elimination the lip
