摘 要:目的探讨大鼠全脑缺血-再灌注损伤后APC-Cdh1蛋白的表达变化。方法雄性SD大鼠60只,体重280~350g,随机分成假手术组和缺血-再灌注组。采用改良Pulsinelli4-VO法建立SD大鼠全脑缺血模型,在损伤后不同时间点,免疫组化染色检测Cdh1的表达部位并且采用Western blotting检测大鼠海马组织APC-Cdh1蛋白的表达。结果免疫组化检测显示APC-Cdh1在海马区及皮层区中均有大量表达。与假手术组相比,缺血-再灌注组术后第1天APC-Cdh1蛋白表达减少,术后第3天升高(P<0.05),术后第7天又降低。结论APC-Cdh1可能与中枢神经系统损伤有着密切的关系。Objective To investigate the expression of APC-Cdh1 protein after cerebral ischemia-reperfusion injury.Methods 60 male Sprague-Dawley rats were randomly divided into Sham-operated group(SH) and ischemia-reperfusion group(IR). The rats of ischemia-reperfusion groups were induced by four-vessel occlusion (4-VO). At different times after injury,the expression of APC-Cdh1 of rat hippocampus was observed by Western blotting and immunohistochemistry.Results Compared with sham-operated group,the expression of Cdh1 protein significantly decreased 1 day and increased obviously 3 days,but decreased again 7 days after injury in ischemia-reperfusion group. The immuno-staining showed that APC-Cdh1 was highly cerebral cortex and hippocampus in ischemia-reperfusion group. Conclusion APC-Cdh1 may be involved in the central nervous system injury.
