肺癌细胞与成纤维细胞、炎症细胞相互作用对MMP-1,-2,-9表达的影响

摘　　要：目的:研究单个核细胞、胚肺成纤维细胞和肺癌H460细胞三维立体混合培养对MMP-1、MMP-2和MMP-9表达的影响,并观察MMPs对胶原的降解作用。方法:活性胶原立体培养分为空白对照组(CG)、H460细胞组(HG)、单个核细胞组(MG)、胚肺成纤维细胞组(FG)、H460与单个核细胞混合组(HMG)、H460与胚肺成纤维细胞混合组(HFG)、三种细胞联合培养组(HMFG)共7组,观察癌细胞的生长状况,用明胶酶谱法测定MMP-2和MMP-9表达,Western blotting法测定MMP-1。结果:各组细胞在立体胶原内生长良好;CG与MG未明显分泌MMP-1、MMP-2和MMP-9,其余各组均分泌MMP-1、MMP-2和MMP-9,但HMFG分泌的基质金属蛋白酶明显多于其他各组。结论:单个核细胞、成纤维细胞和癌细胞混合培养后,细胞间相互作用促进MMPs分泌;基质金属蛋白酶可降解细胞外基质(extraclluar matrix,ECM)和其它间质组织,进而促进肺癌的侵袭和转移。Objective：To observe the effects of co-culture of H460 with mononuclear cells and HFL in three-dimensional（3D） collagen gels on the expressions of matrix metalloproteinase-1,-2,-9（MMP-1,-2,-9）.Methods： Cells cultured in 3D collagen gels were divided into control group（CG）,H460 group（HG）,mononuclear cell group（MG）,fetal lung fibroblast group（FG）,co-culture group of H460 with mononuclear cells（HMG）,co-culture group of H460 with HFL（HFG）,and co-culture group of H460 with mononuclear cells and HFL（HMFG） respectively.The growth of cells was observed under microscope.The expressions of MMP-2,-9 were detected by gelatin zymography,and the expressions of MMP-1 were detected by Western blotting.Results： Cells in collagen gels grew well,and MMP-1,-2,-9 were not detected in CG and MG,but could be obviously seen in HG,FG,HMG,HFG and HMFG,and the levels of MMP-1,-2,-9 in HMFG were significantly higher than those in other 4 groups.Conclusion： More MMPs were secreted in HMFG than in the other 5 groups,and mononuclear cells and HFL in carcinoma tissue may promote the invasion and metastasis of cancer cells;MMPs may promote the invasion and metastasis of cancer cells by degrading collagen and ECM of cancer tissue.
