摘  要:目的探讨低氧诱导因子(HIF)-1α及其靶基因血红素加氧酶(HO)-1在慢性脑缺血致认知功能障碍大鼠皮层中表达变化及意义。方法大鼠双侧颈总动脉永久性阻断法(2VO)建立慢性脑缺血模型,Morris水迷宫测定大鼠的学习记忆能力,免疫组化检测大鼠皮层HIF-1α和HO-1蛋白的表达。结果Morris水迷宫表明缺血组大鼠逃避潜伏期和游泳路径均明显延长,免疫组化显示缺血组大鼠3、6、9w皮层HIF-1α、HO-1表达明显高于假手术组(P<0.05),HIF-1α、HO-1表达分别于3、6w达高峰。结论HIF-1α及其靶基因HO-1的高表达,可能作为保护性因素参与慢性脑缺血致认知功能障碍的过程。Objective To investigate the expressive changes and significance of hypoxia inducible factor (HIF)-1α and hemeoxygenase (HO)-1 in the cortex of rats with cognitive impairment induced by chronic cerebral ischemia.Methods The chronic cerebral ischemia model was made by permanent bilateral occlusion of both common carotid arteries (CCA) occlusion (2VO).The rats were evaluated on learning-memory ability by Morris water maze test.The dynamic expressions of HIF-1α and HO-1 in cortex were measured by immunohistochemistry.Results The latency in each trial and swim distance of the 2VO were longer than those of the sham operation.The expressions of HIF-1α and HO-1 in the cortex of rats were higher than those of the sham operation group.The peaks of HIF-1α and HO-1 protein were respectively at the third and sixth week in the ischemia group.The difference between two groups was significant (P〈0.05).Conclusions Both HIF-1α and HO-1 are highly expressed,may be involved in the course of cognitive impairment of chronic cerebral ischemia.
