摘  要:目的:探讨外胚间充质干细胞(Ectomesenchymal stem cells,EMSC)向骨骼肌细胞分化的能力。方法:利用二甲基亚砜(Dimethyl sulphoxide,DMSO)液诱导外胚间充质干细胞分化,通过免疫组化和逆转录聚合酶链式反应(Reverse transcription-polymerase chain reaction,RT-PCR)鉴定,以大鼠成纤维细胞作为对照。结果:RT-PCR结果说明诱导的外胚间充质干细胞表达骨骼肌细胞特异标志(Desmin)。而对照组细胞形态未出现变化,也无骨骼肌细胞标志表达。结论:外胚间充质干细胞经DMSO液诱导后可成为骨骼肌细胞。AIM:To induce the differentiation of ectomesenchymal stem cells (EMSC) to muscle cells. METHODS:The EMSC were cultured in DMEM containing 5 μL/mL Dimethyl sulphoxide(DMSO). Reverse transcription-polymerase chain reaction(RT-PCR) and immunohistochemistry were used to identify the characteristics of the cells. The rat fibroblast was used as control. RESULTS: RT- PCR showed that after 7d induction, EMSCs expressed Myod mRNA. After 14d induction positive desmin staining was detect in EMSCs by IHC. The controll rat fibroblast did not express Myod mRNA and desmin after induction. CONCLUSION: The results indicated that the EMSC can be differentiated into muscle cells by the action of DMSO.
