摘 要:目的:研究人牙周病牙龈组织环氧合酶(COX-2)和诱导型一氧化氮合酶(iNOS)的表达变化。方法:取牙周炎病人牙龈组织15例,牙龈炎病人牙龈组织15例、正畸或第三磨牙拔除时的正常龈组织14例,制作切片进行COX-2和iNOS免疫组织化学染色,观察比较牙龈组织中COX-2、iNOS的表达变化。结果:①COX-2在正常牙龈组织中几乎不表达,在牙龈炎、牙周炎龈组织中均有阳性表达。其中牙周炎组最高,与正常组相比有显著性差异(P<0.05),但与牙龈炎组相差不显著(P>0.05),牙龈炎组与正常组相比亦无显著性差异(P>0.05);②iNOS在正常牙龈组织中几乎不表达,在牙龈炎和牙周炎龈组织中均有阳性表达。其中正常组最低,与牙龈炎组和牙周炎组相比均有显著性差异(P<0.05),而牙龈炎组与牙周炎组相比则无显著性差异(P>0.05)。结论:牙周病牙龈组织中COX-2、iNOS表达升高,可能成为牙周病时组织破坏、牙槽骨吸收的影响因素之一。AIM:To evaluate the cyclooxgenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) expression in periodontal tissues. METHODS:Fifteen patients with chronic periodotitis (CP),15 patients with gingivitis (G) and 15 health controls (H) were included in this study. Gingival tissues were surgically excised. COX-2 and iNOS expression were detected with immunohistochemistry staining. RESULTS:COX-2 and iNOS were not detected in healthy gingival tissues. Positive staining of COX-2 and very strong positive staining of iNOS were detected in the ginvival tissues of CP and G groups. The expression of COX-2 in CP group was significantly higher than that in H group (P〈0.05). No significant difference of COX-2 expression was found between the CP and G group, and also between the G and H group (P〉0.05). The expression of iNOS in CP and G group was significantly higher than that in H group (P〈0.05). No significant difference of iNOS expression was found between the CP and G groups (P〉0.05). CONCLUSION: COX-2 and iNOS expression ware elevated in the inflamed gingival tissues of gingivitis and chronic periodontitis. COX-2 and iNOS may play important roles in the destruction of periodontal tissues.
