摘 要:目的研究奈达铂(NDP)联合顺铂(DDP)对人食管癌细胞株Eca-109的增殖及凋亡的影响,并探讨其机制。方法采用MTT法检测NDP联合DDP对Eca-109细胞的增殖抑制率,中效原理法判断联合用药的相互作用;采用RT-PCR和蛋白印迹法分析Ki-67及Bax表达的变化。结果NDP、DDP均明显抑制Eca-109细胞的生长,且呈剂量依赖性;两者联合使用低浓度时呈拮抗效应,高浓度时呈协同效应;IC50浓度的NDP、DDP和1/2IC50(NDP)+1/2IC50(DDP)对Eca-109细胞的抑制率分别为(41.9±4.1)%、(47.4±2.9)%和(52.5±0.9)%;与单独用药组比较,联合用药组Ki-67基因(蛋白)表达显著降低,而Bax基因(蛋白)表达显著增高。结论1/2IC50浓度NDP和DDP联合应用与两药IC50浓度单独应用相比,对Eca-109细胞增殖的抑制作用和诱导凋亡能力均有明显增强。Objective To investigate the effect of combined use of nedaplatin(NDP)and cisplatin(DDP)on the proliferation and apoptosis of human esophageal carcinoma cells.Methods The cultured human esophageal cancer Eca-109 cells were treated with NDP and DDP.The inhibitory effects of NDP and DDP on the proliferation of Eca-109 cells were evaluated by MTT assay.The median-effect principle was used to analyze the mechanism of the inhibitory effect after the cells were exposed to the combined treatment of NDP and DDP.The changes of Bax,Ki-67 in mRNA and protein level were quantified by RT-PCR and Western blotting,respectively.Results Both NDP and DDP significantly inhibited the growth of Eca-109 cells in a dose-dependent manner.The combination of NDP and DDP presented synergistic effect at high concentrations and antagonistic effect at low concentrations.The inhibition rate of NDP at IC50,DDP at IC50 and combined NDP and DDP at half IC50 concentrations were(41.9±4.1)%,(47.4±2.9)% and(52.5±0.9)%,respectively.Compared to NDP or DDP treated groups,the expression of Ki-67 mRNA and protein in combination group was decreased;but the expression of Bax mRNA and protein was increased.Conclusion Compared with NDP or DDP alone,combination of NDP and DDP shows more effective in inhibition of proliferation and induction of apoptosis of Eca-109 cell line.
