摘 要:目的:探讨HBV与AFB1协同致肝癌作用机制。方法:用免疫组化观察53只树鼠句肝及肝癌组织IGF-Ⅱ、HBxAg、p21及p53蛋白表达。结果:接受HBV和AFB1的树鼠句肝癌诱发率和IGF-Ⅱ、HBxAg、p21检出率分别为52.9%、82.4%、52.9%及29.4%;只感染HBV的分别为11.1%、22.2%、11.1%和11.1%;仅摄入AFB1的分别为15.8%、26.3%及15.8%(p21);空白对照无肝癌及上述基因蛋白表达。带瘤肝IGF-Ⅱ、HBxAg及p21检出率均显著高于无瘤者,而表达这3种基因蛋白的动物肝癌发生率也均明显高于阴性者。突变的p53蛋白表达仅见于7例(58.3%)中低分化的肝癌。结论:HBV与AFB1共同激发这些基因的异常表达可能是其协同致肝癌机制之一。Purpose To study the mechanism of synergistic effect in hepatocarcinogenesis induced by HBV infection and aflatoxin B 1 intake. Methods Immunohistochemical staining was used in formalin fixed, paraffin embedded sections of cancer and liver tissues. The significance of the data was statistical evaluate by χ 2 test. Results The incidence of hepatocellular carcinoma(HCC) was 52 9% in experimental tree shrews received both HBV and aflatoxin B1 (AFB 1). It was significantly higher than that of animals exposed to HBV (11 1%,Group B), or aflatoxin B1(15 8%, Group C) alone. HCC was not found in control animals (Group D). The expressions of insulin like growth factor Ⅱ(IGF Ⅱ) were 82 4%,22 2%,26 3% and 0 in Group A, B, C and D respectively. The significant differences of IGF Ⅱ were observed between group A and B, C and D ( P <0 05). The expressions of p21 were 29 4%, 11 1%,15 8% and 0 in group A, B, C and D respectively. The positivity rate of HBxAg was significantly higher in Group A than that in Group B (52 9% vs 11 1%, P <0 05). The parallel relations between the incidence of HCC and the over expressions of these genes proteins have been found in each group. On the other hand, the expressions of these genes in tumor bearing tree shrews were significantly higher than that in no tumor bearing animals. The expression of p53 mutation was 58 3% in HCC of tree shrews. Conclusion These foundings suggest a synergistic effect of HBV and AFB 1 in activation of these genes in tree shrews. Over expressions of these genes may play an important role in the courses of hepatocarcinogenesis in tree shrews.
