摘 要:在生理酸度条件下(pH 7.4),以溴化乙锭(EB)为荧光探针,运用荧光光谱法并结合DNA熔点、黏度和盐效应等实验手段,研究了毒死蜱与DNA的相互作用。结果表明,随着毒死蜱的加入,EB-DNA体系的荧光强度逐渐减小,表明毒死蜱能够与EB竞争结合DNA,将EB-DNA体系中的EB置换出来;同时毒死蜱的加入使DNA的熔点升高、粘度增大,上述实验现象表明,毒死蜱以嵌插方式作用于DNA的亲核位点,意味着毒死蜱进入生物体后有可能通过形成DNA加合物的形式造成DNA损伤。The interaction between chlorpyrifos and calf thymus DNA in physiological buffer(pH 7.4) was investigated by fluorescence spectroscopy combined with melting point effect of DNA,viscosity and salt effect using ethidium bromide(EB) as a fluorescence probe,respectively.The results showed that fluorescence of EB-DNA complex was quenched with addition of chlorpyrifos,indicating that chlorpyrifos can strongly compete with EB to bind DNA and displace EB from EB-DNA system.Moreover,the relative viscosity and value of melting temperature of DNA increased in the presence of chlorpyrifos.Based on the above results,it could be inferred that the binding of chlorpyrifos with DNA was an intercalation binding,the chlorpyrifos can bind to base pairs of DNA to produce DNA adducts.The chlorpyrifos might bring the chemical trauma to DNA of the organisms.
