摘  要:目的 通过观察姜黄素(Cur)对碱性成纤维细胞生长因子(bFGF)诱导人乳腺癌细胞(MDA-MB-231)增殖的影响,探讨Cur治疗乳腺癌的作用机制.方法 将人乳腺癌细胞MDA-MB-231用含10%胎生血清的RPMI-1640培养基培养.同时根据加入Cur的不同浓度将实验分为7组:空白对照组、bFGF组(bFGF 5 ng/ml)、干预1组(bFGF 5ng/ml+Cur 1 μmol/L)、干预2组(bFGF 5ng/mI+Cur 2.5 μmol/L)、干预3组(bFGF 5 ng/ml+Cur 5 μmol/L)、干预4组(bFGF 5ng/ml+Cur 10 μmol/L)、干预5组(bFGF 5 ng/ml+Cur 20 μmol/L),并在倒置显微镜下观察各组细胞形态,通过MTT法检测Cur对细胞增殖的影响.结果 bFGF组可显著促进人乳腺癌细胞(MDA-MB-231)的增殖,与对照组的差异有统计学意义(P〈0.05),且其作用随着处理时间的延长而增强.各干预组均可显著抑制人乳腺癌细胞(MDA-MB-231)的增殖,且呈时间和浓度依赖性.结论 Cur可有效抑制bFGF诱导的人乳腺癌细胞增殖,可能为其用于乳腺癌治疗的机制所在.Objective To investigate the effects of curcumin on proliferation of human breast carcinoma cells induced by bFGF. Methods Cultured breast carcinoma MDA-MB-231 cells were divided into 7 groups: the blank control group, bFGF group (5 ng/ml of bFGF), intervention group 1 (5 ng/ml of bFGF + 1 p tooL/L of curcumin), intervention group 2 (5 ng/ml of bFGF + 2.5 p tooL/L of curcumin), intervention group 3 (5 ng/ml of bFGF + 5 p tooL/L of curcumin), intervention group 4 (5 ng/ml of bFGF + 10 p tooL/L of curcumin) and intervention group 5 (5 ng/ml of bFGF + 20 p tooL/L of curcumin). The morphological changes of cells in each group were observed with inverted microscope and the cell proliferation was determined by MTT assay. Results The proliferation of MDA-MB-231 cells was induced significantly by 5 ng/mL of bFGF compared with the blank control group (P〈0.05) and this effect was enhanced with time extension. However, the proliferation effect on MDA-MB-231 cells was inhibited significantly by curcumin in a time-and concentration-dependent manner. Conclusion Curcumin inhibits the proliferation of human breast carcinoma cells induced by bFGF, which is associated with the cell apoptosis.
