摘 要:目的建立人膀胱癌细胞BIU-87的高侵袭力亚系,并对侵袭过程进行观察。方法通过构建侵袭小室,以穿透人工基底膜、聚碳酸脂膜能力为依据筛选具有体外高侵袭能力的膀胱癌细胞亚系。运用扫描电镜观察细胞侵袭的过程,通过生长曲线、细胞周期、体外侵袭力对母代与子代细胞体外增殖及侵袭能力进行对比。结果通过扫描电镜观察到了细胞的侵袭运动。筛选后的各子代细胞,经16代以上传代后,细胞形态与母代无明显变化,细胞增殖速度较母代有明显加快(P<0.05),体外侵袭能力较母系明显提高(P<0.01)。结论运用改良的侵袭实验筛选膀胱癌细胞系的高侵袭力亚系的方法是可靠的,建立的高侵袭力亚系细胞为今后对膀胱癌侵袭及转移的研究提供了较为可靠的细胞模型。Objective To establish the high invasive subseries cell lines of bladder neoplasm strains of BIU-87,and to observe the invasive process in vitro.Methods By using a specially constructed invasive chamber,the cells that have penetrated the matrigel and polycarbonate adipose membrane were selected and cultured to establish offspring cell lines with high invasive power.We had also observed the process of cell's invasion in vitro by scanning electron microscope,and the growth curve,cell cycle and invasive power of the perfusion has been compared with those of their original cells of three bladder neoplasm strains.Results After over 16 generation,the morphous of the subseries cells was similar to that of their original cell lines.The in vitro invasiveness was elevated significantly(P0.01),and the propagation speed was obviously accelerated compared with their origins(P0.05).Conclusion The method of constructing subseries cell lines of bladder neoplasm strains is reliable,and the subseries cell lines can be used as cell models for studies of bladder neoplasm's invasiveness and metastasis.
