摘 要:目的:研究minTBP-PRGDN双靶向融合肽与金属钛表面结合的稳定性及其影响因素。方法:采用FITC标记三种多肽(minTBP-PRGDN、minTBP和PRGDN),在不同离子强度、孵育温度及pH值条件下将多肽溶液分别与钛片孵育过夜,利用荧光显微镜比较分析三种多肽在不同条件下与钛片的结合强度。结果:三种多肽与钛表面均存在结合力,其结合力受离子强度、孵育温度和pH的影响。合适的离子强度有助于多肽在钛表面的结合,在一定范围内,多肽的钛结合能力随着温度的递增而存在一个递减的过程,pH=7.0是多肽与钛表面结合的最适宜pH。结论:minTBP-RGD融合肽不影响minTBP与钛表面的结合能力,4℃、pH=7.0是多肽与钛表面结合适宜环境条件。Objective:To explore the stability of a chimeric peptide of minTBP-PRGDN when binding to titanium surface under different incubating condition.Method:Three peptides of minTBP-PRGDn,minTBP and PRGDN sequence were synthesized and labled with FITC.After overnight incubation with titanium disks under different environmental ion strength,temperature and pH,the stability of peptides was investigated by fluorescence microscope.Result:The binding ability of peptides to the surface of titanium was modulated by ion strength,temperature and pH.Suitable environmental ion strength helped to improve the binding ability of peptides.The binding ability decreaced as the temperature increased.It was suitable for peptides affinity at pH =7.0.Conclusion:The conjunction of minTBP and PRGDN did not influence the stability of minTBP on titanium surface.It is the most suitable eniviromental condition for peptide binding to tiantium surface in 4 ℃ and pH=7.0.
