摘 要:目的:研究急性髓性白血病病人外周血中肿瘤抑制基因RIZ1的表达状况和该基因启动子区甲基化状态及两者之间的关系。方法:以37例初发急性髓性白血病病人和15例正常人外周血标本为研究对象。RT-PCR检测RIZ1 mRNA水平,甲基化特异性PCR(MSP)检测RIZ1基因启动子区甲基化状态。结果:急性髓性白血病病人标本与正常人标本比较,RIZ1基因的表达下降,差异具有统计学意义(P<0.05)。急性髓性白血病标本中RIZ1基因启动子区甲基化率为29.7%(11/37)。存在甲基化的急性髓性白血病病人外周血标本中,RIZ1基因表达缺失或降低。结论:RIZ1基因表达下降与急性髓性白血病的发生有关,其部分机制在于基因启动子区甲基化。Objective: To investigate the expression of RIZ1 gene in acute myeloid leukemia and to analyze the relationship between this alteration and the promoter hyper-methylation of RIZ1 gene.Methods: The expression of RIZ1 mRNA in peripheral blood from 37 patients of primary acute myeloid leukemia samples and in peripheral blood from 15 normal person's samples was detected by reverse transcripition-polymerase chain reaction(RT-PCR).Methylation-specific PCR(MSP)was used to assay the methylation of RIZ1 gene promotor region.Results: Compared with normal person,the RIZ1 gene expression level decreased significantly in acute myeloid leukemia(P0.05).The Methylation rate was 29.7%(11/37) in acute myeloid leukemia.The expression of RIZ1 mRNA was found to be lower in the acute myeloid leukemia samples with RIZ1 promoter methylation.Conclusion: Down-regulation of RIZ1 gene expression plays an important role in the development of acute myeloid leukemia,and this alteration is partially caused by RIZ1 gene promoter methylation.
