摘  要:目的观察复健片在不同时间点对脑梗死大鼠脑组织Nogo-A表达的影响,探讨其促进神经发生的作用机制。方法240只SD大鼠随机分为正常对照组、假手术组、模型组和药物组共四个大组,并根据脑梗死病程每组又分为3d、1周、2周、4周、6周共5个亚组,每个亚组动物数为12只。用改良的LongaEZ法制备大脑中动脉阻塞大鼠模型,药物组灌胃给予复健片水溶液,余各组分别灌胃给予同剂量的蒸馏水。用免疫组织化学法观察模型大鼠梗死周边区脑组织Nogo-A的表达。结果模型组大鼠脑内的Nogo-A在梗死后即开始表达,逐渐增高,2周时达到高峰,第6周时还处于高表达状态,明显高于正常对照组。3d时药物组中Nogo-A的表达与模型组比较差异无显著性(P>0.05),其余各药物组中Nogo-A表达与模型组比较均有非常显著性差异(P<0.01)。结论复健片可抑制Nogo-A的表达,从而促进神经再生。Objective To observe the effect of a traditional Chinese nedicine,"Fujian tablet",on the expression of Nogo-A in brain tissue of rats with cerebral infarction at the different stages,and to explore its mechanism on neural regeneration.Methods Two hundred and forty healthy male Sprague-Dawley rats were randomly divided into 4 groups:normal control,sham operation,model and medicine groups,which were randomly divided into five subsets(n = 12) at 3 d,1 w,2 w,4 w and 6 w stages after cerebral infarction,respectively.The rat models with middle cerebral artery occlusion were successfully established by an improved Longa EZ procedure.Fujian tablet was dissolved in water and given to the rats of treatment groups by gastric gavage,while the other groups received distilled water.The expression of Nogo-A in the brain tissue around the infarction was detected by immunohistochemistry.Results Nogo-A expression was observed at a short period after cerebral infarction in the model group,and increased gradually,reached a peak at 2 weeks and was still positive at 6 weeks after infarction.The differences between model and normal control groups were significant at the same stage(P 0.01).The difference between medicine group and model group was not significant(P 0.05) at 3 days, but significant(P 0.01) at other stages.Conclusion The Fujian tablet can promote neurogenesis by inhibiting the expression of Nogo-A.
