摘 要:目的通过载银纳米二氧化钛义齿基托材料体外细胞毒性的测定,初步评价载银纳米二氧化钛的生物安全性。方法采用噻唑蓝(MTT)比色法检测载银纳米二氧化钛义齿基托材料(载银纳米二氧化钛浓度30g/L)100%、50%浸提液培养小鼠成纤维细胞L-929细胞2、4、7d后吸光度值(OD值),并计算细胞相对增殖率,以6级毒性分类法评级,采用单因素方差分析法对各实验组及对照组的OD值进行统计学分析,显微镜观察细胞的生长状况。结果实验组各观察期L-929细胞生长良好,形态正常;载银纳米二氧化钛抗菌剂(<30g/L)的义齿基托的细胞毒级别为0级。结论载银纳米二氧化钛抗菌剂的义齿基托无细胞毒性。Objective To investigate the cytotoxicity of poly methyl methacrylate denture base with Ag-embedded nano-TiO2 in vitro,in order to evaluate the biocompatibility of materials primarily.Methods Colorimetric MTT(tetrazolium) assay was used,mouse lung fibroblast L-929 were cultured in the 50%,100%extraction of denture base with Ag-embedded nano-TiO2 (30 g/L) for 2,4,7 d respectively.Light absorption values (OD) were assured with enzyme -linked immunoassay instrument,cell relative growth rate (RGR) were calculated.The cytotoxicity was evaluated with 6 grade classification and OD values were analyzed statistically by ANOVA.Cell growth status was observed under microscope.Results The L-929 cells showed normal proliferation and morphology during observation periods.The cytotoxicity of denture base with Ag-embedded nano-TiO2 (30 g/L) was 0 grade.Conclusion There was no cytotoxicity in the denture base with Ag-embedded nano-TiO2.
