干扰素α-2b对兔眼角膜毒性的实验研究

摘　　要：为评价青光眼滤过性手术后应用干扰素α－２ｂ预防和治疗滤过道的纤维疤痕化对眼组织的毒性作用，我们对兔眼前房内直接注入干扰素α－２ｂ以及兔眼行巩膜咬切术后滤过区旁结膜下注射药物后进行角膜组织的检查：角膜上皮用１％荧光素钠染色，角膜内皮用锥兰和茜素红进行内皮活性染色，并对全层角膜组织结构作光镜检查。结果发现：抗青光眼术后滤过区旁结膜下注射干扰素α－２ｂ５×１０５ＩＵ／０．２ｍｌ连续３～１４天，注射总量１．５×１０６ＩＵ～６×１０６ＩＵ，以及前房内直接注入干扰素α－２ｂ１×１０６ＩＵ２４小时和第１０天后，角膜上皮、内皮以及全层组织结构无变化。说明兔眼对干扰素α－２ｂ具有良好的耐受性。n　order　to　evaluate　the　corneal　toxicity　of　adminstered　interferonalpha　2b(IFN　α2b)　as　an　antifibrotic　treatment　after　glaucoma　filtering　surgery,We　investigated　the　corneal　changes　following　subconjunctivally　applied　IFN　α2b　5×105IU　at　filtering　bleb　for　3—14　days　after　sclerectomy　in　white　rabbits　and　directly　injection　IFN　α2b　1×106IU　into　anterior　chamber.Our　results　showed　that　no　sign　of　toxicity　on　corneal　epithelium　by　application　of　1%　sodium　fluorescein　and　no　evidence　of　corneal　endothelium　toxicity　by　viability　examination　with　dual　staining　of　typan　blue　and　alizanin　red　S.We　also　did　not　observe　any　histologic　changes　of　corneal　structure　by　light　microscopy.We　believed　that　rabbits　eyes　seem　to　be　welltolerate　with　high　doses　of　interferon　by　local　used,and　subconjunctivally　injection　IFN　α2b　at　filtering　bleb　after　glaucoma　filtering　surgery　may　be　a　safe　drugapplied　method.
