摘  要:目的:研究双脱甲氧基姜黄素体外抑制SKOV3卵巢癌细胞抑制作用的体外实验研究。方法:(1)将不同浓度双脱甲氧基姜黄素(0、10ug/ml、20ug/ml、40ug/ml)作用于人卵巢癌SKOV3细胞6、12、24h,采用四甲基偶氮唑盐(MTT)比色法检测SKOV3细胞的增殖活性;(2)流式细胞仪观察双脱甲氧基对SKOV3卵巢癌细胞周期的作用,免疫组化检测增殖细胞核抗原(PCNA)的表达。结果:(1)双脱甲氧基姜黄素对SKOV3细胞具有增殖抑制作用,且具有浓度和时间依赖效应;(2)双脱甲氧基姜黄素能诱导SKOV3细胞G1期阻滞,通过抑制细胞生长周期,降低细胞增生速率抑制肿瘤细胞增殖;(3)双脱甲氧基姜黄素能下调SKOV3细胞PCNA蛋白表达,并呈一定的浓度依赖性。结论:双脱甲氧基姜黄素能明显抑制人卵巢癌SKOV3细胞的体外增殖,可能为临床治疗卵巢癌提供了一种新的方法。Objcetive: To investigate the effect and mechanism (bisdemethoxycurcumin,BDC inhibiting SKOV3 cells in vitro. DC)ofdifferent concent rations (0,10,20 and 40ug/ml ) was administered to SKOV3 cells of ovarian cancer for 6h, 12h and 24 h, respectively. The proliferation of SKOV3 cells was measured by MTT assay.2. Induction of apoptosis was detected with FCM. 3.Protein expressions of PCNA were detected with immunohistochemistry; Results: (1) (bisdemethoxyeurcumin,BDC) inhibited the growth of SKOV3 with a concentration and time-dependent effect.(2) BDC inhibited SKOV3 cells in GI phase and inhibited the proliferation of SKOV3 cells by blocking the growth of the cell cycle and reducing the cell growth rate. (3)BDC inhibited the expression of PCNA in SKOV3 cells and inhibited with a concentration-dependent effect. Conclusion: (bisdemethoxycureumin, BDC)earl significantly inhibit the proliferation of SKOV3 cells in vitro. BDC could provide a novel method in the clinical therapy of or ovarian cancer.
