摘 要:采用ITSPCR-RFIt方法对5个野生木耳属菌株进行分析,并以7个黑木耳栽培菌株和1个毛木耳菌株为参照。通过6个限制性内切酶消化共获得20条清晰易辨的多态性DNA条带,使用NTSYspe2.1生物软件对DNA条带信息进行分析,结果显示:13个木耳属菌株的遗传相似系数变异范围为0.500—1.000,遗传相似系数变化较大;13个供试菌株在相似系数为0.547时聚在一起,在0.721时聚为3个类群;实验结果支持Auhm、Au5931和Au59641为黑木耳属菌株;Audx菌株鉴定为毛木耳属菌株。研究结果表明ITSPCR—RFLP技术可为区别黑木耳和毛木耳菌株提供重要信息。5 wild Auricularia strains were studied using internal transcribed spacer (ITS) PCR-RFLP method with 7 cultivated Auricularia auricula and 1 Aur/cu/ar/a polyrricha strains as control. Results showed there were,in total ,20 distinct and polymorphic DNA bands after di- gested by 6 restriction endonucleases respectively. The dendregram was generated by unweighted pairgroup method with arithmetic average (UPGMA) method with NTSYSpc2.1 bio-software. Results showed: (i) The similarity coefficients of 13 Auricuhtria Strains ranged from 0. 108 to 0. 822; (ii) All strains were clustered together at the similarity coefficient of 0. 547 ,and clustered into 3 groups at the similarity coefficient of 0.721 ; ( iii ) Accessions of Auhm, Au5931 and Au59641 were considered as Auricularia auricular strains, while accession of Audx was identified as Auricuiarla polytricha strain. And ITS PCR-RFLP technique proved a vital role to play in distinguishing between Auricuiarla auricular and Auricularia polytricha strains.
