摘 要:植物在长期进化过程中形成了自我防御机制,能够产生特异的抗性蛋白来应对各种生物因子的胁迫,其中茉莉酸信号途径是植物必需的防御机制。本研究对拟南芥施用茉莉酸处理和对照叶片蛋白质的差异表达变化,并试图通过蛋白质组学技术来检测茉莉酸诱导的蛋白质,通过蛋白质双向电泳发现有28个蛋白点发生了显著的变化,其中19个蛋白点上调表达,9个蛋白点下调表达。选择10个差异蛋白点进行了质谱鉴定,它们分别是苏氨酸蛋白激酶、热激蛋白、不依赖于维生素B12的蛋氨酸合酶、腺嘌呤琥珀酸合酶、碳酸酐酶等,这些蛋白质可能在拟南芥叶片应答茉莉酸反应的过程中起到重要作用,进一步进行差异蛋白的功能分析对揭示茉莉酸信号通路和植物防御机制的关系具有重要的作用。Plants produce jasmonic acid or methyl jasmonate (MeJA) in response to many biotic stresses, particu- larly insect herbivores. Jasmonate is essential signal pathway for insect defense in plants. To get a better understanding the defense response in plants for MeJA,two-dimensional electrophoresis(2-DE)accompanied with mass spectrometry (MS) were therefore applied to the study. In this study, we identified some proteins involved in jasmonate signaling, which is essential for plant defense. Of the experiments,28 protein spots with at least 2-fold changed expression were found in Arabidopsis,10 out of which were successfully identified using MALDI-TOF/TOF MS analysis. The MeJA re- sponsive proteins included serine-threonine protein kinase,chloroplast heat shock protein 70-1 ,independent VB12 me- thionine synthetase,adenylosuccinate synthase,carbonic anhydrase, and et al. We were able to determine a few pro- teins that displayed abundance changes in response to MeJA. Understanding how the plant response to MeJA is regulated will provide tools for a better study of induced defense mechanism.
