摘  要:Arf(ADP-ribosylation factors)作为小G蛋白家族中的一类,在植物细胞的胞内运输中起着重要的作用。从采后2d的香蕉(Musa accuminata L.AAA group)果实cDNA文库中筛选到一条Arf基因,命名为MaArf1。序列分析表明,MaArf1在植物中是高度保守的。与水稻、苜蓿、马铃薯、小麦和拟南芥等物种中Arf基因的相似性都超过95%。并且MaArf1分别在24-31(序列为GLDAAGKT,Arf蛋白P结构域),67-71(序列为DVGGQ,G'结构域),126-130(序列为NKQDL,G结构域)的氨基酸处也具有保守的GTP结合结构域。根据已获得的cDNA序列设计引物,研究又获得了该基因的基因组序列。对其结构分析发现,该序列全长1998bp,由5个外显子和4个内含子组成。内含子序列中存在大量的重复序列,并且在第4个内含子中有一个完整的开放阅读框,推测这些序列可能参与基因的表达调控,也可能与RNA水平上的可变剪接有关。ADP-ribosylation factors (Arf), a family of small GTP-binding proteins, play important roles in intracellular translocation in plants. A homolog of Arfwas isolated from banana (Musa accuminata L. AAA group) by screening cDNA library from postharvest banana fruit and was designated as MaArfl. Sequence analysis of deduced amino acids showed that Ma-ARF1 is highly conserved compared with ARF proteins from other plants. Very high similarity (with over 95% identity) was found between Ma-Arf1 and those from rice (OsArf), alfalfa (MtArf), potato (StArf), wheat(TaArf) and Arabidopsis (AtArf1). Several conserved motifs in Arf proteins were also observed. Three consensus sequences proposed to be involved in GTP binding were present at amino acid positions 24-31 (sequence GLDAAGKT, P motif of A rfprotein), 67-71 (sequence DVGGQ, G' motif DXXGQ), and 126-130(sequence NKQDL, G motif NKXD). The putative genomic sequence of MaArf1 was also amplicated by a pair of primers designed according to the cDNA. Analysis of its structural characteristic showed that the gene was 1 998 bp in size, including 5 exons and 4 introns. A number of repeat sequences were found in introns, while a complete open reading frame was present in intron 4, which was proposed to be involved in the regulation of gene expression.
