摘  要:从健康鸡的肠道中分离、鉴定并筛选出1株耐酸和耐胆盐能力较强的乳杆菌,命名为乳杆菌L3株。采用单因素法和正交试验设计法对乳杆菌L3株的培养条件进行了优化,结果表明,乳杆菌L3株的最佳液体发酵培养基组成是4%的蔗糖、0.4%的胰蛋白胨、10%的番茄汁、0.5%β-磷酸甘油二钠、0.2%的柠檬酸铵、0.5%的乙酸钠、0.1%的吐温-80、0.1%的L-Cys盐酸盐、0.05%的MgSO4、0.02%的MnSO4,最适接种时间是16-18h,最适培养条件是温度40℃、培养时间28 h、初始pH 5.0,在此培养条件下,乳杆菌L3株的菌落总数可达8.14×10^8cfu/mL。Isolated,identified and screened out from the healthy chickens a Lactobacillus with strong tolerance of acid and bile salt,and named as Lactobacillus strains L3.Optimized the culture conditions of Lactobacillus strains L3 by Using single-factor method and orthogonal experimental design method.The results showed that best liquid fermentation medium composition of Lactobacillus strains L3 were as follows: sucrose 4%,tryptone 0.4%,tomato juice 10%,β-glycerophosphate disodium 0.5%,ammonium citrate 0.2%,sodium 0.5%,tween-80 0.1%,L-Cys hydrochloride 0.1%,MgSO4 0.05%,MnSO4 0.02%,the optimal vaccination time is 16-18h,the optimal culture conditions of temperature is 40 ℃,incubation time 28h,the initial pH5.0.In this culture conditions,the total colony of Lactobacillus strains L3 is 8.14×10^8cfu/mL.
