摘 要:目的探讨血管生成素-2(Ang-2)和环氧化酶-2(COX-2)在膀胱移行细胞癌中的表达及其与膀胱移行细胞癌病理分级、临床分期之间的关系,以及两者表达之间的相互关系。方法选取经临床病理诊断的膀胱移行细胞癌标本60例,正常膀胱组织20例。应用免疫组化染色方法检测Ang-2和COX-2在膀胱移行细胞癌中的表达情况。结果Ang-2和COX-2在正常膀胱组织中均无表达;在膀胱移行细胞癌标本中均呈过表达。膀胱移行细胞癌中Ang-2和COX-2的阳性表达率分别为47.7%(28/60)、55%(33/60)。膀胱移行细胞癌中,Ang-2和COX-2阳性表达率随其病理分级和临床分期增加而升高。经统计检验,Ang-2与COX-2的等级相关系数rs为0.541,差异有统计学意义(P<0.01)。结论膀胱组织在癌变过程中获得了Ang-2、COX-2受体。Ang-2、COX-2的阳性表达率均随膀胱移行细胞癌的病理分级、临床分期增加而升高,表明Ang-2、COX-2阳性率越高,膀胱移行细胞癌的恶性度越高,浸润越深。膀胱移行细胞癌中Ang-2和COX-2的表达具有两两正相关关系;两者相互作用,通过影响肿瘤血管形成,共同参与膀胱移行细胞癌的发生、发展。Objective The study is to investigate the relationships among the expression of angiopoietin-2 (Ang- 2), Cyelooxygenase-2 (COX-2) and the clinicopathological variables in patients with transitional cell carcinoma (TCC) of urinary bladder. Methods The expression of Ang -2 and COX-2 were assessed by immunohistochemistry in tissue sections from 60 patients with transitional cell carcinoma of the urinary bladder and in 20 normal bladder samples. Results There is no marked expression of Ang-2 and COX-2 in normal bladder samples. The positive expression of Ang-2 and COX-2 was detected in TCC samples. The positive rate of Ang-2 and COX-2 was 47.7%(28/60) and 55%(33/60), respectively. In TCC, positive Ang-2 and COX-2 expression was significantly correlated with histological grade, clinical stage. There were relatively correlations in overexpression of Ang-2 and COX-2 in TCC of the urinary bladder (rs= 0. 541, P〈0. 01). Conclusion These results suggest that bladder tissue gets Ang-2 and COX-2 receptor in eancerization. The overexpression of Ang-2 and COX-2 is associated with tumour grades and stages. These findings suggest that Ang-2 and COX-2 may play an important role in development and progression of TCC.
