摘 要:从鳞翅目亚洲玉米螟(Ostrinia funacalis(Guenée))3龄幼虫提取基因组DNA,通过PCR扩增和测序,获得其核糖体小亚基18S rRNA基因(18S rDNA)的序列,利用C lustalW与六足总纲中22个目(纲)昆虫的18S rRNA基因序列进行多重联配.结果表明,昆虫18S rRNA有4段序列较为保守,其中第2区段最为保守.分别用全长序列和第2保守区段构建分子系统发育树,结果显示:保守区段构建的系统发育关系比较符合传统分类,所列各目(纲)中毛翅目与鳞翅目亲缘关系最近,昆虫纲中的衣鱼目、蜉蝣目、蜻蜓目与弹尾纲、双尾纲、原尾纲距离较近,属于比较古老的昆虫类群.18S rRNA sequence was amplified from genomic DNA of Ostriniafunacalis Guenée by using PCR method in this study. Alignments of Ostrinia funacalis Guenée 18S rRNA with other 22 orders were further carried out. The results indicat that 18S rDNA contains four conserved domains and the second domain is the most conserved. Systemic tree analysis suggested that the evolution relation consists with traditionally taxonomic results when using the most conserved 18S rDNA domain to construct phylogenetic tree. The phylogenetic tree also suggested that Trichopter and Lepidoptera were clustered together and Ephemeropterodae and Odonata and Zygentoma have closer relationship with Collembola, Diplura and Protura, which belong to an old insect genus.
