摘 要:目的:探讨紫杉醇对胆管癌QBC939细胞作用的敏感性并研究紫杉醇对胆管癌细胞周期及细胞凋亡率的影响,并初步探讨紫杉醇诱导胆管癌细胞发生凋亡的机制,为胆管癌的临床治疗寻找新的治疗药物。方法:体外培养胆管癌细胞QBC939,采用不同浓度紫杉醇予以干预,通过细胞形态观察和MTT实验,初步研究紫杉醇对人胆管癌细胞QBC939的敏感性;应用流式细胞仪检测紫杉醇诱导胆管癌细胞凋亡,同时对细胞周期的变化和动力学予以检测。结果:各浓度紫杉醇对人胆管癌细胞QBC939生长均有明显抑制作用;胆管癌细胞被紫杉醇明显阻滞在S期及G2/M期,且抑制作用呈剂量和时间依赖效应。结论:紫杉醇对人胆管癌细胞QBC939增殖的抑制作用呈剂量和时间依赖效应;紫杉醇能诱导人胆管癌QBC939细胞发生凋亡。Objective :To investigate the sensibility of cholangiocarcinoma QBC939 cell lines to paclitaxel and the changes in cell cycle and the mechanism of apoptosis of cholangiocarcinoma cell induced by paclitaxel. Methods: QBC939 cell lines were cultured in vitro, paelitaxel was applied to interfere the growth of QBC939 with different con- centration, to investigate the sensibility of cholangioearcinoma cell ( QBC939 ) to paclitaxel by observing cell morphous and MTT method ; Flow cytometry was applied fo detect the changes of cell cycle and cytodynamics of cholangiocarci- noma cell induced by paclitaxel. Results: Paclitaxel could inhibit the growth of eholangioeareinoma QBC939 cell lines obviously at different concentration;Cells had been main hold -uped in S phase and G2/M phase. Conclusion:Pacli- taxel can inhibit the growth of the QBC939 and the inhibitory effect is time - centration dependent; QBC939 cell lines can be induced apoptosis by paclitaxel.
