摘  要:目的:探讨HER-2RNA干扰(RNAi)对HER-2阳性乳腺癌细胞的化疗敏感性的影响。方法:构建能够表达HER-2siRNA的质粒载体,利用载体转染HER-2阳性乳腺癌细胞SKBR-3,RT-PCR与蛋白质印迹法检测HER-2mRNA与蛋白表达,受转染的细胞与不同浓度的化疗药物表柔比星共培养,MTT法检测细胞活性。结果:SKBR-3细胞HER-2基因受到RNAi后,HER-2mRNA及蛋白表达分别下降约60%与55%,细胞活性出现明显下降,肿瘤细胞对表柔比星的半数抑制浓度(IC50)为0.25μg/μL,相较对照组而言,化疗敏感性提高明显。结论:乳腺癌HER-2是理想的RNA干扰治疗靶点,靶向HER-2的RNAi可以抑制肿瘤细胞生长,而且能够增加肿瘤细胞化疗敏感性。OBJECTIVE:To study the effects HER-2 gene RNA interference (RNAi) on the chemo-sensitivity of HER-2-positive breast cancer cell. METHODS:A plasmid vector capable of mediating HER-2 RNAi was constructed,and HER-2 positive breast cancer cell line SKBR-3 was transfected with constructed vector. The expression of HER-2 mRNA and protein was analyzed by RT-PCR and Western blotting; Also,transfected cells were co-cultured with different concentrations of epirubicin and the growth of SKBR-3 cells was analyzed by MTT. RESULTS:The expression of HER-2 mRNA and HER-2 protein was downregulated about 60% and 55%,respectively,in response to vector-mediated HER-2 RNAi. Furthermore,the chemo-sensitivity to epirubicin of the SKBR-3 cell was enhanced significantly by HER-2 gene RNAi and the IC50 was 0.25 μg/μL. CONCLUSION:HER-2 is a good target for RNAi and RNAi,targeting HER-2 gene can cause HER-2 positive breast cancer cell growth inhibition and increasing sensitivity to chemotherapeutics.
