摘  要:目的:探讨磷酸化组蛋白(γH2AX)的表达水平,预测肝癌细胞对于电离辐射的敏感性。方法:以不同剂量X射线照射肝癌细胞系HepG2.215,流式细胞术检测肝癌细胞中表达γH2AX的细胞分数;免疫细胞化学法检测肝癌细胞中γH2AX灶点数,MTT比色法检测放射线照射后的细胞活性。结果:肿瘤细胞经X射线照射后,γH2AX的表达呈剂量依赖性,随照射剂量的增高而增高;γH2AX的表达水平与细胞死亡率呈正相关(r2=0.984,P<0.05),即γH2AX的表达越高,细胞的死亡率越高。结论:γH2AX表达水平预测肝癌细胞对于放射线治疗的敏感性有较高的可行性和可靠性。OBJECTIVE:To study the sensitivity of hepatic carcinoma cell HepG2.215 to ionizing radiation by using phosphorylated H2AX(γH2AX) as a predictor. METHODS:HepG2. 215 cells were exposed to X-rays of different doses respectively,The proportion of HepG2.215 cells expressing γH2AX was measured by flow cytometry,the number of γH2AX foci in HepG2.215 cells was measured by immunocytochemistry,and the cell proliferation was measured by MTT. RESULTS:After HepG2.215 cells were exposed to X-rays,the expression of γH2AX was dose-dependent and increased linearly with the doses; The level of γH2AX was positively correlated with cell mortality (r^2=0.984,P0.05). The more the expression of γH2AX was,the higher cell mortality was. CONCLUSION:It is feasible and reliable to use γH2AX for predicting the sensitivity of HepG2.215 cells to ionizing radiation agents.
