摘  要:目的探讨低氧环境下血管生成素1(Ang-1)对人胃癌细胞中整合素β1的影响。方法利用腺病毒作为载体将已构建成功含Ang-1基因的重组腺病毒(Ad-Ang-1)瞬时转染人胃癌细胞株BGC-823,利用低氧诱导剂氯化钴进行低氧干预低氧转染(D)组,以单纯转染(B)组及单纯低氧干预(C)组作为对照,以未转染未行低氧干预的正常细胞作为空白对照(A)组,利用半定量RT-PCR,免疫细胞化学和Western blot方法对4组细胞中整合素β1的mRNA和蛋白表达水平进行分析。结果D组整合素β1的mRNA及蛋白表达明显高于A、B、C组(P<0.05)。结论低氧环境下转染Ang-1能够明显提高人胃癌细胞BGC-823中整合素β1mRNA及蛋白的表达,表明低氧环境下Ang-1促进肿瘤细胞的侵袭转移。Objective To evaluate the effects of angiopoietin-1 on the expression of integrin β1 in human gastric cancer cell line BGC-823 under hypoxia.Methods The BGC-823 cells were divided into 4 groups of transfected with Ad-Ang-1 transiently and induced by hypoxia mimic cobalt chloride(group D),transfected with Ad-Ang-1 alone(group B),induced by hypoxia mimic cobalt chloride alone(group C),and blank control(group A).The expressions of integrin β1 mRNA and protein were detected by RT-PCR,immunohistochemistry and Western blot.Results The BGC-823 cells were transfected successfully.The mRNA and protein expressions of integrin β1 in group D were significantly higher than those in groups of A,B and C(P0.05).Conclusion Hypoxia and transfection of Ang-1 into human gastric cancer cell line BGC-823 can significantly increase the expression of integrin β1,indicating that Ang-1 under hypoxia promotes the invasion and metastasis of the tumor cells.
