摘　　要：目的研究环氧化酶2(COX-2)抑制剂NS-398对胃癌培养细胞系BGC-823增殖及凋亡的影响。方法应用MTT法检测NS-398对胃癌细胞BGC-823细胞增殖的影响;应用流式细胞术、逆转录聚合酶链反应(RT-PCR)和Westernblot法检测NS-398对胃癌细胞BGC-823细胞凋亡的影响。结果NS-398随剂量的增加及作用时间延长对胃癌细胞呈抑制作用;体外NS-398能减少BGC-823细胞株COX-2的表达,对BGC-823有细胞毒作用,可增加细胞凋亡率。结论NS-398可抑制胃癌BGC-823细胞的增殖,促进胃癌BGC-823细胞的凋亡。其可能机制是与抑制COX-2表达有关。Objective To study the effect of selective COX-2 inhlbitor NS398 on gastric cancer cell proliferation and apoptosis.Methods Gastric cancer cell lines were treated with NS-398.Proliferation of the cells was detected by MTT assay,apoptosis of the cells was measured by flow cytometry,RT-PCR and Western blotting.Results NS-398 inhibited the growth of gastric cancer in dose-and time-dependent manner.NS-398 in vetro suppressed the proliferation of BGC-823 and inhibited cycolooxygenase 2（COX-2） expression.NS-398 had cytotoxic effect on the cell lines and induced apoptosis.Conclusion NS-398 can inhibit the cell proliferation and induce cell apoptosis of gastric cancer,which may be related to its effect of inhibiting COX-2 expression.
