摘  要:目的研究慢病毒MED19-shRNA包装颗粒感染胃癌SGC-7901细胞后对其细胞增殖和细胞周期的影响。方法应用MED19-shRNA-pLVTHM慢病毒载体感染胃癌SGC-7901细胞,采用RT-PCR和Western blot的方法检测MED19基因的沉默效率,通过MTT和克隆形成实验研究MED19基因在SGC-7901细胞增殖中的作用,并用流式细胞技术试验检测MED19基因沉默后对细胞周期的影响。结果成功构建的MED19-shRNA慢病毒载体并使MED19基因沉默成功,感染SGC-7901细胞后细胞增殖能力显著降低、细胞克隆形成能力明显减弱、细胞周期阻滞于S期。结论MED19基因沉默后对胃癌SGC-7901细胞增殖和细胞周期均有显著影响。Objective To observe the changes of cell proliferation and cell cycle of SGC-7901 cells after infected by MED19-shRNA packaging lentivirus.Methods Western blot and Real-time PCR were used to detect the silence efficiency of MED19 in the infected SGC-7901 cells by MED19-shRNA-pLVTHM lentiviral vector,then MTT assay and colony formation assay were used to detect the effect of MED19 on cell proliferation,and FCM technology was applied to test the changes of cell cycle after the gene MED19 silence.Results The recombinant MED19-shRNA lentiviral expressing vector was successfully constructed and the MED19 gene was silenced efficiently. The cell proliferation and colony numbers were decreased by MED19 expression after infected by SGC-7901 cells,and the cell cycle was arrested in phase S by MED19 silence. Conclusion The silence of MED19 gene can obvious affect the cell proliferation and cell cycle in SGC-7901 cells.
