摘  要:目的建立一个火药烟雾对体外人支气管上皮细胞影响的实验平台。方法自行设计制作气体发生及气体与细胞作用装置,将培养的人支气管上皮细胞直接与原始状态下火药烟雾接触,以不同剂量火药烟雾及不同处理时间对支气管上皮细胞活性及代谢的影响为指标,观察实验平台使用效果。结果本实验平台内能够观察到火药烟雾致支气管上皮细胞损伤的剂量、时间效应关系,平台本身对细胞没有影响,实现了原始气体与细胞的直接作用,所设计烟雾发生装置及烟雾与细胞作用平台可进行火药烟雾的毒理学分析。结论实验平台结构简单,成本低廉,能够成功检测气态物质的细胞水平毒理作用,有助于减少实验动物的使用,可广泛用于气态物质的毒理学分析。Objective To establish the platform for evaluating the effect of gunpowder smoke on cultured bronchial epithelial cells.Methods A gas generating device was designed for the interaction between gas and cells.Cultured human bronchial epithelial cells were directly contacted with the gunpowder smoke in its original state.Different amounts of gunpowder smoke and treatment time were used as an indicator to observe the effect of gas in the device on the activity and metabolism of bronchial epithelial cells.Results The gas generating device could induce bronchial epithelial cell injury in a dose-and time-dependent manner while the device itself had no effect on the cells,suggesting that the original gas in the smoke generating device can directly interact with the cells and the device can be used in analysis of gunpowder inhalation toxicology.Conclusion The experimental device,simple in structure and low in cost,can detect the toxicological effect of gas on the activity and metabolism of bronchial epithelial cells at cellular level,reduce the use of laboratory animals,and can thus be widely used in toxicological analysis of gas.
