摘 要:目的:探索通过高温环境、醋酸铅致敏和静脉注射脂多糖的综合方法制作大鼠温病暑热证模型的方法。方法:人工气候箱制造高温环境,麻醉动物,颈总动脉置入导管并连接生理多导仪,尾静脉注射醋酸铅致敏,1/2h后尾静脉注射脂多糖,动态观察肛温、脉搏、呼吸、收缩压、舒张压和脉压差,检测炎症介质IL-1β、IL-2、IL-4、IL-6、IL-10、IL-18、TNF-α,并进行肝、肺病理检查。结果:模型组的脉搏较对照组增快,肛温、呼吸、收缩压、舒张压和脉压差无组间差别;模型组的IL-1β、IL-4、IL-6、IL-10、TNF-α高于对照组,差别有统计学意义;模型组在4、5、6h的死亡率高于对照组,差别有统计学意义。结论:成功建立了大鼠温病暑热证模型,该模型病情比较严重,具有明显的炎症特点和中医暑热证的特点。Objective: Endue summer heat syndrome rat model of epidemic febrile disease by high temperature environment,plumbi acetas infusion and lipopolysaccharide(LPS)infusion through vein.Methods: Rats were raised in high temperature environment made by artificial climate box before experiment.Pipes were inserted into arteria carotis communis of anesthetized rats and connected with physiological multiplying channel instrument.Then,rats received plumbi acetas infusion and lipopolysaccharide infusion 1/2 hour later through vena caudalis.Rectal temperature,arterial pulse,respiration,contractive pressure,diastolic pressure and pulse pressure of rats were observed,while IL-1β,IL-2,IL-4,IL-6,IL-10,IL-18 and TNF-α were tested by enzyme linked immunosorbent assay.Samples of lung and liver were got for pathological section at the end of experiment.Results: The arterial pulse of the model group was statistically faster than the control group while there was no statistical significance between groups in rectal temperature,respiration,contractive pressure,diastolic pressure and pulse pressure.The level of IL-1β,IL-4,IL-6,IL-10,TNF-αin the model group were statistically higher than the control group.There was statistical difference in mortality which was higher in the model group.Conclusion: Summer heat syndrome rat model of epidemic febrile disease was endued successfully.There was serious pathogenetic condition,obvious inflammation and character of summer heat syndrome in this model.
