摘  要:目的建立稳定可靠的线栓法大鼠大脑中动脉阻塞(MCAO)永久性脑缺血模型。方法对Koizumi和ZeaLonga模型构建方法加以改进。40只雄性SD大鼠随机均分为实验组和假手术组,分别进行模型制作和模型缺血的评价,采用2,3,5-氯化三苯四氮唑(2,3,5-Triphenyltetrazolium chloride,TTC)染色来计算大鼠脑梗死体积。结果大鼠MCAO模型制作成功率为95%;神经功能缺失评分1~3级百分率为95%。结论实验所采用的改良术式构建大鼠MCAO模型具有稳定性好、可重复率高、操作简便和手术时间短等优点,为基于大鼠永久性脑缺血模型开展的下游相关实验提供了良好的研究平台。Objective To set up a stabilized and reliable model of rat permanent middle cerebral artery occlusion (pMCAO). Methods Rat pMCAO experiment was performed based on the previous Koizumi and Zea Longa's models with significant modification. Forty male Sprague-Dawley (SD) rats were randomly divided into experimental group and sham-operated group (n=20 in each group),and occurrence of rat cerebral ischemia in each group was assessed. In this study,2,3,5-Triphenyltetrazolium chloride(TTC) staining were carried out to detect the infarct area in the brain of experimental rats. Results In experimental group,the successful ratio of cerebral ischemia model was 95%,and neurological deficit scores of 1~3 grade was 95%. Conclusion This modified pMCAO model is an ideal model with better stabilization,higher reproducibility,easier execution and simpler operation when compared with the previous one.
