脆性组氨酸三联体基因在宫颈癌中的表达及其与细胞凋亡的相关性

摘　　要：目的探讨脆性组氨酸三联体(FHIT)基因在宫颈癌中的表达情况及与细胞凋亡的相关性。方法利用免疫组织化学技术检测63例宫颈癌和20例慢性宫颈炎中FHIT蛋白的表达情况,并用脱氧核糖核苷酸末端转移酶介导的缺口末端标记技术(TUNEL)检测宫颈癌细胞凋亡情况。结果(1)FHIT阳性表达率宫颈癌为28.60%,显著低于慢性宫颈炎的70%(P<0.01)。(2)宫颈癌中FHIT蛋白表达减弱与年龄、临床分期无关(P>0.05);而与宫颈癌组织学分级、淋巴结转移有关(P<0.05)。(3)随FHIT蛋白表达缺失,细胞凋亡指数逐渐降低。结论FHIT基因异常表达与宫颈癌的潜在恶性生物学行为密切相关,可作为评价预后的指标之一,并且参与了肿瘤细胞凋亡的调节。Objective To explore the clinical importance of change in gene expression,we assessed the level of FHIT gene in benign lesions and carcinomas of the cervix.Methods The expression of FHIT protein was detected by immuohistochemistry SP in 20 chronic cervicitis and 63 cervical carcinomas.The apoptosis index（AI）was studied by TUNEL in cervical carcinomas.Results（1）FHIT positive expression rate in cervical cancers was 28.60%,which was significantly lower（70%）than that in chronic cervicitis（P0.01）.（2）There was no statistical differances between FHIT protein expression with age and clinical stage（P0.05）.But,reduced FHIT protein expression was significantly associated with histological grades and lymph node metastasis（P0.05）.（3）The mean AI was lower in cervical carcinoma with reduced FHIT expression than those with normal FHIT protein expression（P0.05）.Conclusion These findings suggest that the tumor suppressor FHIT gene may present a molecular target in highly aggressive phenotypes of cervical carcinoma,and play an important role for induction of apoptosis of human cervical carcinoma.
