摘 要:目的:研究Mad4基因与神经再生之间的相关性。方法:通过RT-PCR方法研究Mad4基因在壁虎主要组织中的表达分布与断尾损伤中的变化,以及运用原位杂交技术分析Mad4基因在脊髓中的表达定位。结果:原位杂交结果显示,多疣壁虎脊髓中Mad4基因的阳性信号主要存在于灰质及室管膜细胞;Mad4基因在脑、脊髓、卵巢、肝、肺、肾、肌肉和睾丸组织中均有表达,各自表达的丰度有一定的差异,其中在脑、脊髓和睾丸中的表达量最高。在断尾损伤中,多疣壁虎Mad4基因表达呈动态变化,脊髓损伤近侧端3天时表达量下降,随后1周和2周时表达量恢复到正常水平;脊髓损伤远侧端1天和3天时表达量急剧上升,但2周时表达量下降。结论:Mad4基因与壁虎的脊髓再生可能存在一定的相关性。Objective:To study the relation between Mad4 gene and spinal cord regeneration.Methods:Total RNA was extracted from the tissues of gecko,and the first chain of cDNA was obtained by reverse transcription.Mad4 was amplified by PCR.The expression of gecko Mad4 gene was analyzed by reverse transcription polymerase chain reaction(RT-PCR) and in situ hybridization(ISH).Results:Section in situ hybridization demonstrated that the Mad4 gene transcripts in the cervico-lumbar spinal cord were distributed in the gray matter and ependymal cells.Mad4 gene was expressed in all the tissues detected,i.e.the brain,spinal cord,ovary,liver,lung,kidney,muscle and testis.But the expression abundance revealed difference.And the expression abundance of Mad4 gene in the brain,spinal cord and testis was higher.The expression levels of Mad4 gene in the distal spinal cord showed that Mad4 gene decreased at 3 days and went back to the control level at 1 week and 2 weeks after amputation.For the proximal spinal cord,the expression levels of Mad4 gene markedly increased at 1 day and 3 days after amputation,but decreased at 2 weeks.Conclusion:Mad4 gene is possibly involved in the spinal cord regeneration.
