摘　　要：目的：观察蛋白和核酸水平Bcl-2结合抗凋亡基因1（Bcl-2-Associated Athanogene1,BAG-1）在多发性骨髓瘤（multiple myeloma,MM）的表达,探讨BAG-1在多发性骨髓瘤发病机制中的作用及其临床意义,为多发性骨髓瘤的治疗寻找相关的理论依据。方法：选取皖南医学院附属弋矶山医院2006年10月-2008年10月初诊MM患者骨髓活检标本28例用于BAG-1免疫组化检测;同时抽取上述患者骨髓液提取单个核细胞,用于BAG-1mRNA RT-PCR检测。结果：初诊多发性骨髓瘤患者的BAG-1蛋白表达水平率为64.29%,和对照组相比增高明显（P=0.008）。同样在核酸水平,BAG-1mRNA在初诊骨髓瘤患者表达率为71.4%,高于对照组（P=0.007）。BAG-1高表达的MM肿瘤负荷高、与多项临床指标有关。结论：BAG-1蛋白及BAG-1基因在多发性骨髓瘤患者中高表达,与对照组相比有显著性差异,与临床多项指标有关,经分析可能参与多发性骨髓瘤的发生,BAG-1蛋白及其基因高表达可能提示患者不良预后。Objective：To investigate the role of Bcl-2-associated athanogene（BAG-1）expression in the pathogenesis of multiple myeloma（MM）and its clinical significance for providing theoretical evidence in treatment of this disease.Methods：The bone marrow biopsy specimens were taken from 28 patients newly diagnosed as MM in the affiliated hospital of Wannan Medical University between Oct.2006 and 2008 for the immunohistochemical detections of BAG-1,and mononuclear cells were obtained from the bone marrow extract of the patients described above for detecting BAG-1 mRNA by reverse transcription polymerase chain reaction（RT-PCR）.Results：BAG-1 protein was positively expressed in 64.29% of the newly diagnosed MM cases,which was significantly higher as compared with the control subjects（P=0.008）.Higher BAG-1 mRNA expression was also seen in the newly diagnosed MM patients regarding nucleic acid level.The positive rate accounted for 71.4% and was higher than that of the controls（P=0.007）.Conclusion：Higher BAG-1 expression in multiple myeloma correlates with a series of clinical indicators,suggesting that BAG-1 may be involved in the development of MM and poor prognosis.
