摘 要:目的:观察视觉发育可塑期后(P120)单眼斜视(monocular strabismus,MS)大鼠视皮质神经元c-fos蛋白表达的情况,探讨斜视性弱视模型成年大鼠视皮质内视觉可塑性存留状态的差异,为临床防治大龄儿童斜视性弱视提供参考依据。方法:13日龄(P13)SD大鼠共16只,随机分为正常组(N)和单眼斜视组(MS),对视觉发育关键期13日龄(Postnatal,P13)前的幼龄大鼠制作单眼斜视动物模型。斜视组45日龄(P45)做F-VEP(flash visual evokedpotential,闪光视觉诱发电位,F-VEP)检测确立弱视模型,斜视组100日龄(P100)取材,取材前经黑箱饲养24 h,曝光0.5 h处理后灌注固定切取两组大鼠脑组织视皮质,应用免疫组化染色比较MS组与正常大鼠视皮质光刺激诱导c-fos蛋白表达的阳性神经元的差异。结果:①P45MS组大鼠术眼与对侧健眼及正常组F-VEP结果比较:N1P1、P1N2波振幅在术眼表现较大幅下降,差异有统计学意义(P〈0.05);②MS组视皮质17区(OclM、OclB区)Ⅱ-Ⅲ层c-fos蛋白免疫阳性神经元密度高于正常组。结论:视觉发育关键期(P13)内的视觉紊乱(MS)可导致大鼠发生弱视;单眼斜视成年大鼠视皮质神经元经光刺激诱导c-fos蛋白表达增加,提示单眼斜视性弱视成年大鼠视皮质内存留一定程度视觉可塑性。Aim:To explore the persistence of neuronal plasticity of monocular strabismus(MS) amblyopia rats which has overgrown(P120) the critical period of visual development by investigating the expression of c-fos protein in the visual cortex using immuno-histochemistry. Methods: Sixteen healthy SD rats aged 13 postnatal days(Postnatal13,P13) were randomly divided into two groups: normal(N);and monocular strabismus(MS).Operation were enforced to the amblyopia rats to set up the monocular strabismus animal model.Rats with all monocular strabismus were detected by flash visual evoked potential(F-VEP)to be amblyopia animal models.All rats were reared together until postnatal day 100 and lived in dark box 24 h before taking samples.After half an hour exposure to ambient light,all the rats were perfused.Sections of the OclM,OclB areas of visual cortex were cut and then processed for c-fos immunohistochemistry.The differentiation of morphological parameter of neurons in visual cortex together with results of c-fos immunohistochemistry between the amblyopia group and the normal group were analysed statistically.Results: ①Amplitude of N1P1 and P1N2 descend obviously in the strabismus eye of the model rats.②Under light microscope,the density of c-fos immunoreactive cells in OclM、OclB in layers Ⅱ and Ⅲ of visual cortex of adult monocular strabismus amblyopia rats was much higher than that of healthy rats(P〈0.05).Conclusion:As one of the functional identification mark,the expression of c-fos protein in the neurons of visual cortex of amblyopia rats exceed,suggesting there are certain persistence of neuronal plasticity in the visual cortex of monocular strabismus rats which has overgrown the critical period of visual development.
