摘 要:目的通过裸鼠体内试验探讨GRIM-19对子宫颈癌细胞株HeLa的侵袭、凋亡的影响。方法将HeLa/Con-trol、HeLa/GRIM-19接种到裸鼠皮下的移植瘤剥离,采用western Blot检测MMP-2、MMP-9基因的mRNA和蛋白水平表达情况;利用Tumel实验检测细胞凋亡。结果移植瘤的蛋白检测发现MMP-2、MMP-9基因的表达均下降,HeLa/GRIM-19移植瘤中凋亡细胞增多。结论 GRIM-19的表达可以抑制子宫颈癌细胞的侵袭、促进细胞凋亡,可能是子宫颈癌治疗的新靶点。Objective To explore the effects of expression of GRIM-19 on the invasion and apoptosis of cervical cancer in vivo.Methods Tumor tissue of the null mice was stripped after injection HeLa/Control、HeLa/GRIM-19 into the skin for 7 weeks.The protein level of gene expression was investigated by western Blot.Cell apoptosis was examined by Tunel assay.Results The growth of HeLa/GRIM-19 implanted tumor slowed down significantly(P0.01).The protein expression of MMP-2 and MMP-9 was decreased in tumors bearing HeLa/GRIM-19.Apoptosis cells were increased in HeLa/GRIM-19 implanted tumor.Conclusion The expression of GRIM-19 can slow down the invasion of cervical cancer cells,and contribute to cell apoptosis.Suggesting that GRIM-19 may be a new target for the treatment of cervical cancer.
