摘  要:目的:观察不同氧环境下中药罗勒多糖对人卵巢癌细胞株SKOV3生物学行为的影响,探讨其抗卵巢癌作用的可能机制。方法:罗勒多糖分别在常氧(21%O2、5%CO2)和乏氧(1%O2、5%CO2和94%N2)环境中作用于人卵巢癌细胞株SKOV3,形态学观察不同氧环境下各组细胞形态差异;细胞凋亡-Hoechst33258染色检测药物对人卵巢癌细胞凋亡情况的影响;MTT法检测各组细胞增殖能力的变化。结果:与对照组相比,乏氧环境下罗勒多糖能够促进人卵巢癌细胞株SKOV3的凋亡,抑制SKOV3细胞增殖,而常氧环境下罗勒多糖作用相反。结论:罗勒多糖在常氧及乏氧环境下对人卵巢癌细胞株SKOV3生物学行为的影响效应截然相反,提示抗肿瘤药物及生物疗法的体外机制研究设计应充分考虑氧环境的影响。Objective:To investigate the effect of basil polysaccharide on biological behavior of human ovarian cancer cell lines SKOV3 under hypoxia and normoxia ,and explore the potential mechanism of anti-ovarian cancer.Methods:The human ovarian cancer cell lines SKOV3 cultured with basil polysaccharide under normoxia(21%O2,5%CO2)and hypoxia(1%O2,5%CO2,94%N2)respectively.Morphology of different groups of this cell line under different oxygen concentration were observed with inverted phase contrast microscope;the apoptosis of human ovarian cancer cell line were tested by cell apoptosis-Hoechst 33258 staining;the proliferation of the cell line was evaluated by MTT assay respectively.Results:Compared with the control group,basil polysaccharide could promote the apoptosis of human ovarian cancer cell line(SKOV3) and inhibit the proliferation of SKOV3 cell line,but in normoxia the function of basil polysaccharide was opposite.Conclusion:Basil polysaccharide has opposite influence on biological behavior of human ovarian cancer cell lines SKOV3 in vitro under hypoxia and normoxia conditions,indicating that in vitro study on anti-tumor drugs and bio-therapy should take the oxygen environment into consideration.
