整合素αvβ6、MMP-9在46例卵巢上皮癌表达及意义

摘　　要：[目的]探讨卵巢上皮癌中整合素αvβ6、基质金属蛋白酶-9表达水平及临床意义。[方法]临床资料完整的卵巢上皮癌冷冻组织46例及正常卵巢组织10例作为对照。采用反转录—聚合酶链反应(RT-PCR)方法和免疫组织化学法检测αvβ6、MMP-9表达水平。[结果]①卵巢上皮癌中整合素αvβ6mRNA表达强度(0.895±0.189)显著性高于正常卵巢组织(0)(P=0.000);MMP-9mRNA表达强度(0.360±0.126)显著性高于正常卵巢组织(0.031±0.02)(P<0.001)。②卵巢上皮癌中整合素αvβ6阳性表达率为100%,而10例正常卵巢组织均阴性表达(P<0.05)。整合素αvβ6高表达与临床分期、病情进展、肿瘤部位密切相关(P<0.05),与年龄、病理类型、组织学分级、残留灶大小无关(P>0.05)。卵巢上皮癌中MMP-9阳性表达率为92.7%,正常卵巢组织阳性表达率为80%(P=0.000)。MMP-9高表达与临床分期、病情进展密切相关(P<0.05),与年龄、病理类型、组织学分级、残留灶大小、肿瘤部位无关(P>0.05)。③卵巢上皮癌中MMP-9的表达与αvβ6的表达呈正相关(r=0.801,P=0.000)。[结论]上皮性卵巢癌组织中,整合素αvβ6与MMP-9表达均上调,其表达上调与卵巢上皮癌侵袭性和不良预后有关。[Purpose] To investigate the expression of integrin alphavbeta6 （αvβ6） and matrix metalloproteinase-9（MMP-9） in ovarian carcinoma and its significance. [Methods] Tissue samples from 46 cases with ovarian carcinoma and from 10 cases with normal ovarian as control group were studied. The expression of integrin αvβ6 and MMP-9 were detected by RT-PCR method and immunohistochemistry. [Results] （1）The expression intensity of integrin αvβ6 （0.895 ±0.189） and MMP-9 （0.36±0.126） in ovarian carcinoma were higher than those in normal ovary （0 and 0.031±0.020 respectively）（P〈0.001）. （2）The positive expression of integrin αvβ6 in ovarian carcinoma and normal ovarian was 100% and 0 （P〈0.05）.The positive rate in ovarian epithelial carcinoma was related to clinical stage, disease progress and tumor site （P〈0.05）, and not related to age, pathological type, grade and residual tumor volume （P〉0.05）. The positive expression of MMP-9 in ovarian carcinoma and normal ovary was 92.7% and 80% respectively （P=0.000）. The positive rate in ovarian carcinoma was related to clinical stage and disease progress （P〉0.05）, not related to age, pathological type, grade, residual site and tumor （P〈0.05）. （3）There was a significant positive correlation between the expressions of integrin αvβ6 and MMP-9 （r=0.801,P 0.001）. [Conclusion] αvβ6 and MMP-9 are over expression in ovarian carcinoma. They may relate to cancer aggressiveness and poor prognosis.
