摘  要:目的:探讨感染人轮状病毒乳鼠小肠黏膜白细胞黏附分子-1(ICAM-1)、γ-干扰素(IFN-γ)变化的意义及嗜酸乳杆菌的干预效果。方法:选用ICR乳鼠,分为正常对照组(A组)、轮状病毒感染组(B组)、嗜酸乳杆菌预处理组(C组)和嗜酸乳杆菌治疗组(D组),B、C、D组喂服人轮状病毒Wa株。C、D组另喂服嗜酸乳杆菌进行预处理和治疗干预,于病毒感染后第4天随机处死各组8只乳鼠,留取小肠标本,采用免疫组化SABC方法检测ICAM-1、IFN-γ的表达。结果:B组ICAM-1表达细胞数和灰度为(52.66±6.62)%和108.11±2.42,A组为(38.10±4.43)%和101.97±2.10,B组比A组显著增高(P<0.05);C组为(43.93±4.42)%和101.75±2.28,D组为(43.45±5.26)%和101.42±2.23,均较B组显著降低(均P<0.05),C组、D组与A组比较差异无显著性。B组IFN-γ表达细胞数和灰度为(43.51±31.40)%和127.71±3.36,A组为(37.52±3.21)%和117.71±3.39,B组比A组显著增高(P<0.05);C组为(50.08±4.21)%和136.37±3.40,D组为(49.50±4.05)%和135.90±3.49,均较B组显著升高(均P<0.05)。结论:嗜酸乳杆菌对感染人轮状病毒的乳鼠有免疫保护作用,其机制可能与抑制ICAM-1,增强IFN-γ的表达有关。Objective To explore the expressions of intercellular adhesion molecule 1 (ICAM-1) and interferon-γ (IFN-γ) in small intestinal mucosa in suckling mice infected with human rotavirus and the effect of Lactobacillus acidophilus intervention. Methods ICR suckling mice were randomized to normal control group (group A), rotavirus infection group (group B), rotavirus infection group with Lactobaeillus aeidophilus pretreatment (group C), or rotavirus infection group with Lactobacillus acidophilus treatment (group D). 8 mice in each group were executed randomly 4 days after infection. The expressions of ICAM-1 and IFN-γ in the intestine were detected by immunohistochemistry (SABC assay). Results The positive cell population and gray scale for ICAM-1 expression were markedly higher in group B than in group A [ (52.66 ± 6.62)% vs. (38.10 ± 4.43) % and 108.11 ± 2.42 vs. 101.97 ± 2.10, P 〈 0.05 ]. ICAM-1 expression was lower in groups C and D [ (43.93 ± 4.42)% and 101.75 ± 2.28, (43.45 ±5.26)% and 101.42 ± 2.23 ] than in group B (P 〈 0.05 for all comparisons), but did not differ significantly between group C or D and group A. The positive cell population and gray scale for IFN-γ expression were evidently higher in group B than in group A [(43.51 ± 31.40)% vs. (37.52 ± 3.21)% and 127.71 ± 3.36 vs. 117.71 ± 3.39, P 〈 0.05 ] ; and higher in groups C and D [ (50.08 ± 4.21 ) % and 136.37 ± 3.40, (49.50 ± 4.05 ) % and 135.90±3.49], as compared with group B (P 〈 0.05 for all comparisons). Conclusion Laetobaeillus acidophilus has immune protection for the suckling mouse infected with human rotaviros, whose mechanism may be associated with suppressing ICAM-1 expression and increasing IFN-γ expression.
