电针预处理对大鼠肝脏缺血再灌注损伤中肝细胞凋亡的影响

摘　　要：目的电针预处理,对大鼠肝脏缺血再灌注损伤中肝细胞凋亡的影响。方法24只健康雄性Wistar大鼠,随机分为3组,即假手术组(C)、电针刺预处理+缺血再灌注组(EAP+IR)、单纯缺血再灌注组(IR)。EAP+IR、IR组采用Pringle法建立肝缺血再灌注损伤模型,EAP+IR组于术前电针刺激阳陵泉穴30 min。再灌注6 h后,取部分肝组织用于原位DNA末端标记法(TUNEL)检测肝细胞凋亡率的变化;免疫组化测肝组织中bax和caspase-3的表达;WESTERN-BLOT法对肝脏中bcl-2、bax和caspase-3的表达。结果与C组比较,EAP+IR、IR组各种蛋白表达均增加,肝细胞凋亡率也明显升高。与IR组相比,EAP+IR组中肝细胞的凋亡率明显下降(P<0.05),caspase-3和bax蛋白表达量降低,而bcl-2蛋白则明显上调,免疫组化与WESTERN-BLOT法表现一致。结论电针预处理可以减轻缺血再灌注损伤后肝细胞凋亡。Objective To investigate the effects of Electro-acupuncture preconditioning on hepatocyte apoptosis in rats with hepatic ischemia-reperfusion injury（HIRI）.Methods Twenty-four healthy male Wistar rats were randomly divided into three groups,namely sham operation group（C）,electro-acupuncture preconditioning plus ischemia-reperfusion group（EAP ＋ IR） and ischemia-reperfusion group（IR）.EAP ＋IR and IR model was produced by warm Pringle method,preoperative electrical stimulation at Yanglingquan point 30 min of EAP ＋ IR group.6 hours after reperfusion,getting a portion of liver tissue for the rate of hepatocellular apoptosis was determined using the TdT-mediated d-UTP-biotin nick end labeling（TUNEL） method;bax and casepase-3 protein in hepatocytes were examed by immunohistochemistry.The protein expression of bcl-2、bax and caspase3 in liver was tested by WESTERN-BLOT method.Results compared with c group,the expression of various protein were increased and the rate of liver cell apoptosis was significantly higher.The rate of hepatocellular apoptosis（P0.05）,the level of caspase-3 and bax protein were significantly lower in the EAP＋ IR group than that in the I/R group after reperfusion.whereas the level of bcl-2 protein increased dramatically.Conclusion EA preconditioning reduces ischemia-reperfusion injury of liver cell apoptosis
