塞来昔布对大鼠心脏移植后外周血CD_4^+淋巴细胞的影响

摘  要:目的探讨塞来昔布对大鼠心脏移植后急性排斥反应及外周血CD4+淋巴细胞的影响及其作用机制。方法以SD大鼠为供者,Wistar大鼠为受者,进行腹部异位心脏移植改良的Ono心脏移植术。将实验动物随机分为3个空白对照组及3个实验组,手术后第3、5、7 d测定CD4+细胞总数及线粒体凋亡细胞数,并观察移植心脏的病理变化。结果在应用塞来昔布3 d及5 d时,病理损伤轻于对照组。与对照组相比塞来昔布在3 d表现为促使CD4+T细胞凋亡或抑制其增殖;在实验5 d、7 d时,可抑制CD4+细胞的线粒体凋亡。结论在应用塞来昔布3 d抑制急性排斥反应的发生,可能与促使CD4+细胞凋亡或抑制其增殖有关。应用塞来昔布第7 d时,不能抑制急性排斥反应的发生,可能与抑制CD4+细胞的线粒体凋亡有关。Objective To investigate the effects and mechanism of Celecoxib on CD4+ cell after rat allograft heterotopic heart transplantation.Methods All the rats who received heart transplanted were divided into 6 groups at random.Different groups were given to Celecoxib 50 mg/(kg·d) or 0.9% Sodium Chloride 3.0 mL/(kg·d) from day 0 to day 3,day 5 and day 7.The pathologic lesion,the levels of CD4+ cell and CD4+ cell mitochondrial apoptosis in peripheral blood were measured.Results Celecoxib played a definite anti-acute rejection dependant on time.Celecoxib only decreased acute rejection in 3 days and 5 days,anti-rejection effect did not occur in 7 days.Celecoxib induced CD4+ cell apoptosis in 3 days,repressed CD4+ cell mitochondrial apoptosis in 5 days and 7 days.Conclusion Celecoxib can repress acute rejection only during 5 days after transplantation and anti-rejection effect did not occur in7 days.Celecoxib anti-apoptosis effect can be found in 5 days and 7 days.
