摘 要:为进一步证实菠萝蛋白酶对肿瘤细胞的诱导分化作用,采用形态观察计数、血红蛋白定量测定、发光法测定细胞吞噬能力和Northern原位杂交法观察和检测了菠萝蛋白酶诱导K562细胞分化及其p53基因表达变化.结果显示,菠萝蛋白酶可诱导K562细胞向红系和粒/巨噬系两个方向分化;在菠萝蛋白酶作用下,p53基因于给药后8h转录表达增强,24h达高峰,此后又下降,其时相变化先于分化发生.这些结果提示p53基因在Bromelain诱导K562细胞分化过程中可能起启动分化的重要作用,有关机制尚待深入研究.By using method of cell morphology, hemoglobin detection, cell phagocytosis testing with illumination and Northern blot in situ, the impact of inducing differentiation on K562 cells and expression of p53 gene by Bromelain were further investigated. The results showed that K562 cell could be induced to differentiation into both erythroid and granulocytic/macrophage lineages. During Bromelain treatment, the expression of p53 gene was upregulated from 8 hours to the top of 24 hours, then, downregulated again. These results suggested that p53 might play an important role in triggering K562 cell differentiation induced by Bromelain. However, more detailed work may be needed in future.
