摘  要:目的建立胶原包埋人胃癌微小癌巢体外模型。方法利用组织工程技术,以胶原为支架材料,人胃低分化腺癌BGC-823细胞为种子细胞,构建胶原包埋人胃癌微小癌巢体外模型。倒置显微镜观察模型中肿瘤细胞的生长状态;噻唑蓝(MTT)比色法测定细胞增殖曲线;苏木素-伊红(HE)染色进行组织学观察,并与平面培养模型、胶原铺板培养模型比较。结果胶原包埋人胃癌微小癌巢体外模型中,肿瘤细胞呈立体生长,彼此聚集成球状;能够较长时间维持细胞活性,第7天平面培养模型、胶原铺板培养模型和胶原包埋人胃癌微小癌巢体外模型A496值分别为0.608、0.584和0.728,胶原包埋人胃癌微小癌巢体外模型与其他模型比较差异均有统计学意义(P〈0.05);组织切片可见基质中较多微小癌巢形成。结论胶原包埋人胃癌微小癌巢体外模型能较好地模拟体内肿瘤生长状态,可用于抗肿瘤药物及免疫活性细胞的体外筛选。Objective-To establish the collagen-embedded micro-nests tumor model of human gastric cancer in vitro. Methods The collagen was used as the scaffold and the poorly differentiated adeno- carcinoma of human gastric cancer cell line-BGC-823 cells utilized as the seed cells. Under the inverted phase contrast microscopy, the growth of tumor cells was observed. Cell proliferation was measured by using MTT assay. The histological changes of the tumor model were examined by hematoxylin and eosin (HE) staining. All the aspects were compared with the monolayer culture and the collagen overlay culture. Results Compared with the monolayer culture and the collagen overlay culture, tumor cells in collagen- embedded micro-nests tumor model grew in three-dimensional culture. Cells adhered to each other and formed spheroids. The A496 was 0. 728 at the 7th day, significantly different from that in other two groups (P 〈 0. 05). Many micro-nests were observed histologically in the matrix. Conclusion The collagen-em- bedded micro-nests tumor model can mimic the actual tumor in vivo availably and can be used to screen the antitumor drugs and immune cells conveniently.
